摘要
目的诱变选育普伐他汀高产菌株。方法采用化学诱变剂亚硝基胍(NTG)对出发菌进行诱变处理,以固体培养基初筛、摇瓶培养复筛获得突变株,再对突变株进行遗传稳定性的测定。结果适宜的NTG诱变剂量为浓度0.4mg·mL^(-1)、处理时间30min,获得了一株遗传性状稳定、高产普伐他汀的突变菌株PN-87,其普伐他汀产率较出发菌株提高47%。结论采用亚硝基胍可以诱变选育获得普伐他汀高产菌株。
OBJECTIVE To screen out highly-productive strains of Pravastatin by mutation.METHODS The original strain was treated using chemical mutagens-nitroguanidine(NTG),and then screened by solid medium and re-screened by shake-flask culture.Afterwards,mutant strain was generated.Then the genetic stability of the mutant is determined.RESULTS With an optimal mutagenic dose of NTG at a concentration of 0.4 mg·mL^-1,the strain was treated by 30 mins.A mutant strain PN-87 with stable and high yield of Pravastatin was obtained,representing an increase of 47%in the yield of Pravastatin when compared to that of the starting strain.CONCLUSION Highly-productive strain of Pravastatin can be obtained by mutation using nitroguanidine mutagenesis.
