摘要
随着新型诱导系在育种领域的选育和诱导效率的提高,玉米单倍体加倍技术发展也至关重要,但现有单倍体加倍方法效率普遍较低。为了进一步提高玉米单倍体的加倍效率,探索最佳的加倍条件,采用对玉米籽粒胚部划胚技术及不同质量分数的秋水仙素和不同处理时间对玉米单倍体籽粒进行划胚加倍。结果表明,对玉米单倍体的划胚处理显著高于自然加倍;不同质量分数的秋水仙素及不同处理时间对加倍后植株散粉率等影响显著,其中,秋水仙素质量分数为0.04%、处理时间3 h的散粉率最高,加倍效果最佳。
With the breeding and induction efficiency of new inducing lines in the field of breeding, the development of maize haploid doubling technology is also crucial, but the efficiency of existing haploid doubling methods is generally lower. To further improve the doubling efficiency of maize haploids, explore the best doubling conditions, the paper used the technique of embryo sizing of corn kernels and double the ploidy of maize haploid seeds by using different mass fraction of colchicine and different treatment time. The results showed that the treatment of maize haploid was significantly higher than natural doubling, the colchicine with different mass fractions and different treatment time had significant effects on the powder loosening rate after doubling, when the mass fraction of colchicine was 0.04%, and the treatment time was 3 h, the powder dispersal rale was the highest and the doubling effect was the best.
