ECS对去势小鼠学习记忆障碍作用在Morris水迷宫和神经电生理长时程增强方面的研究 被引量:5

摘要 目的研究锁阳乙酸乙酯提取物(ECS)在Morris水迷宫和神经电生理长时程增强(LTP)方面对去势小鼠学习记忆障碍的作用。方法选用36只SPF级雌性ICR小鼠,将其随机分为对照组、模型组、ECS组,每组12只,模型组、ECS组经7%水合氯醛0. 06 ml/10 g麻醉后摘除双侧卵巢以制备去势小鼠模型,造模成功后,ECS组给予0. 47 mg/ml ECS提取物0. 1 mg/10 g灌胃,对照组和模型组给予等量生理盐水灌胃,1次/d,连续给药8周。8周后对各组小鼠进行Morris水迷宫实验及神经电生理实验。分析小鼠穿越原平台次数与兴奋性突触后场电位(fEPSP)斜率百分比的相关性。结果实验第5天,模型组潜伏期较对照组显著延长,ECS组逃避潜伏期较模型组显著缩短,差异有统计学意义(P<0. 05);模型组穿越原平台次数较对照组显著减少,ECS组穿越原平台次数较模型组显著增加,差异有统计学意义(P<0. 05); HFS刺激后,模型组fEPSP斜率百分比较对照组显著下降,ECS组fEPSP斜率百分比较模型组显著升高,差异有统计学意义(P <0. 05); 36只小鼠穿越原平台次数与fEPSP斜率百分比呈相关性(P <0. 05)。结论 ECS在Morris水迷宫和神经电生理LTP方面对去势小鼠突触传递可塑性及学习记忆能力有明显改善,可为临床防治学习记忆障碍提供理论依据。 Objective To investigate effect of ethyl acetate extract of cynomorium songaricum(ECS)on learning and memory impairment in castrated mice in Morris water maze and long-term potentiation(LTP).Methods 36 SPF female ICR mice were randomly divided into control group,model group and ECS group,with 12 rats in each group.Rats in model group and ECS group were ovariectomized.Corresponding treatments(ECS 0.47 mg/ml for ECS group and NS for model and control group,0.01mg/10g)were given respectively for 8wks.Thereafter,the Morris water maze test and nerve electrophysiology experiment were performed in each group.The correlation between the number of times crossing the original platform and the field excitatory postsynaptic potential(fEPSP)slope were analyzed.Results On the 5th day of the experiment,the escape latency of the model group was significantly longer than control group and ECS group(P<0.05).The number of times crossing the original platform in the model group was significantly lower than in control group and ECS group(P<0.05).The fEPSP slope in model group was significantly lower than in control group and ECS group(P<0.05).The number of times the 36 mice crossing the original platform was correlated with fEPSP slope(P<0.05).Conclusion ECS can significantly improve the synaptic transmission plasticity and learning and memory ability of ovariectomized mice in Morris water maze and LTP,providing theoretical basis for clinical prevention and treatment of learning and memory impairment.
