摘要
建立了超高效液相色谱法测定玉米低聚肽粉中AY含量的方法。样品超声萃取后过滤,WatersAcquityUPLCBEHC18色谱柱(50mm×2.1mm,1.7μm)分离,以0.1%三氟乙酸/乙腈为流动相,流速0.13mL/min,二极管阵列检测器,检测波长223nm。在2.5mg/L^40mg/L范围内呈线性相关,相关系数R为0.9999。样品中加标回收率为96.12%~107.07%,重现性RSD=1.04%。该法快速准确,简便灵敏,分离度高,能够满足玉米低聚肽中AY的检测要求。
A HPLC method for the determination of AY in corn oligopeptide powder was established. The sample was filtered by ultrasonic extraction and separated by the UPLC BEH C18 column (50mm×2.1mm, 1.7mm),Using 0.1%trifluoroacetic acrylonitrile as the flow phase, the flow rate was 0.13mL/min. Detector was diode array detector, detecting wavelength was 223nm. Linear correlation in range of 2.5mg/L~ 40mg/L, correlation coefficient R was 0.9999. The recovery rate was 96.12%~107.07%, and the reproducible RSD=1.04%. The method was fast, accurate, simple and sensitive, with high separation degree, and could meet the requirement of AY detection in corn oligopeptide.
