摘要
对甜橙果树下土壤中分离并保藏于该实验室的一株耐碱酵母进行研究。用通用引物ITS1/ITS4对该菌株基因组DNA进行扩增,并把扩增产物送去测序,然后把该序列用BLASTN进行检索和MEGA7.0软件分析。结果表明,该菌株与KT336517.1同源性大于99%,且位于系统发育树的同一分支上,所以鉴定该菌株为耐碱酵母(Galactomyces);通过对耐碱菌株生活条件的测试,发现该菌株最适合生长温度为28℃,最适合生长的pH为3.7~4.2,同时在酒精浓度为12%和14%,pH在2.5~9均能生长。
In this paper, a alkali-resistant yeast isolated from the soil under the sweet orange tree and preserved in the laboratory was studied. The DNA extract of the alkali-tolerant yeast was amplified by universal primer ITS1/ITS4, and the expanded yield was sent for sequencing, then the sequence was retrieved by BLASTN and MEGA7.0 Software analyzed.the PCR product shared more than 99% homology with KT336517.1 and was located in the same branch of phylogenetic tree with KT336517.1, so it could be identified as alkali-tolerant yeast (Galactomyces). By testing the living conditions of the alkali-resistant strain, it was found that the optimum temperature for growth of the strain was 28℃, and the optimum pH value for growth was 3.7~4.2. while the alcohol concentration is 12%and the 14%,ph value can grow in 2.5~9.
