和胃安神方对失眠模型大鼠血清Th1/Th2平衡及中缝背核GFAP、BDNF表达的影响 被引量:11

Effects of Hewei Anshen decoction on serum Th1/Th2 balance and expression of GFAP and BDNF in dorsal raphe nucleus of model rats with insomnia

摘要 目的探讨和胃安神方对失眠模型大鼠血清Th1/Th2平衡及中缝背核胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)、脑源性神经营养因子(brain derived neurotrophic factor,BDNF)表达的影响。方法 32只雄性Wistar大鼠完全随机法分组。采用对氯苯丙氨酸(para-chlorophenylalanine,PCPA)法进行造模,造模成功后,对各组大鼠进行相应方式的灌胃处理,7天后进行腹主动脉采血和冰上取脑组织保存待测。采用酶联免疫吸附法(Enzyme-Linked ImmunoSorbent Assay,ELISA)检测血清中IFN-γ、TNF-α、IL-4及IL-10,采用免疫组化观察GFAP、BDNF在中缝背核的表达情况。结果与模型组比较,和胃安神方组IFN-γ和TNF-α含量均显著升高( P <0.01)、 IL-10 含量显著降低( P <0.01)、IL-4含量无明显差异( P >0. 05),IFN-γ/IL-4比值显著升高( P <0.01 );与模型组比较,地西泮组、和胃安神方组大鼠中脑中缝背核GFAP 和BDNF平均光密度(mean optical density,MOD)值均显著升高( P<0.05,P<0.01;P<0.01,P <0.05)。结论 GFAP、BDNF的表达增强和Th1/Th2平衡向Th1方向偏移可能是和胃安神方治疗失眠的疗效机制之一。 Objective To investigate the effects of Hewei Anshen decoction on serum Th1/Th2 balance and expression of glial fibrillary acidic protein (GFAP) and brain derived neurotrophic factor (BDNF) in model rats with insomnia. Methods Thirty-two male Wistar rats were grouped randomly. The model was established by para-chlorophenylalanine (PCPA) method. After successful modeling, the rats in each group were given different decoctions through intragastric infusion. After 7 days, their blood was collected from abdominal aorta and the brain tissue was taken on ice for storage. The Serum of IFN-γ, TNF-α,IL-4,IL-10 was examined by enzyme-linked immunosorbent assay(ELISA) and immunohistochemistry(IHC) was used to observe the expression of GFAP and BDNF in the dorsal raphe nucleus. Results Compared with the model group, the levels of IFN-γ and TNF-α in the Hewei Anshen decoction group significantly increased ( P <0.01);the IL-10 levels remarkably decreased ( P <0.01);the IL-4 levels showed no obvious differences ( P >0.05), and the IFN-γ/IL-4 ratio largely increased ( P <0.01 ). Compared with the model group, the mean optical density (MOD) of GFAP and BDNF of the rat dorsal raphe nucleus in the diazepam group and the Hewei Anshen decoction group increased remarkably( P <0.05, P <0.01;P <0.01, P <0.05).Conclusion The enhanced expression of GFAP and BDNF, and the shift of Th1/Th2 balance to Th1 may be one of the therapeutic mechanisms of the Hewei Anshen decoction in treating insomnia.
