慢生根瘤菌接种花生的结瘤能力 被引量:2

Nodulation Ability of Peanut Bradyrhizobium Strains Inoculated on Peanut

摘要 为筛选合适的花生根瘤菌菌株及提高花生产量提供理论依据,选用分离自花生(DASA03005、DASA 03183和DASA 03028)与合萌(ORS 278与STM 6978)的5种慢生根瘤菌,人工接种至花生植株,检测植株结瘤数,采用乙炔还原法测定固氮酶活性;采用BOX-PCR指纹图谱分析技术,比较接种菌株与结瘤菌株的DNA指纹图谱,揭示菌株基因组的差异。结果表明:菌株DASA 03005和ORS 278不能与花生形成结瘤,DASA 03028、DASA 03183和STM 6978能与花生形成结瘤,且接种DASA 03028、DASA03183菌株的花生其单株植株结瘤数和生物量都大于接种STM 6978菌株的;菌株DASA 03028、DASA03183和STM 6978与结瘤菌株的DNA指纹图谱基本一致,菌株DASA 03005和ORS 278则与结瘤菌株的差异较大。接种菌株的结瘤数越多,花生植株的干重越重,固氮酶活性越强,地表植株的生物量也相应较大。 To provide a theoretical basis for screening suitable peanut rhizobium strains and increasing peanut yield,five Bradyrhizobium isolates from peanuts(DASA 03005,DASA 03183 and DASA 03028)and Aeschynomene indica(ORS 278 and STM 6978)were used for artificial inoculation on peanut plants,the number of nodules in the plants was detected,and the nitrogenase activity was determined by acetylene reduction method.The DNA fingerprint of the inoculated strains and the nodulation strains were compared by BOX-PCR fingerprint analysis technique to reveal the differences of genomes of the strains.Results:Strains DASA 03005 and ORS 278 could not form nodules with peanuts,while DASA 03028,DASA 03183 and STM 6978 could form nodules with peanuts,and the per plant number of nodules and biomass of peanuts inoculated with DASA 03028 and DASA 03183 strains were higher than that of STM 6978;The DNA fingerprints of the inoculated strains DASA 03028,DASA 03183 and STM 6978 were basically the same as those of the nodulation strains,while that of DASA 03005 and ORS 278 were greatly different with nodulation strains.The more the number of nodules in the inoculated strain,the higher the dry weight of the plant,the stronger the nitrogenase activity,the larger the biomass of the surface plants.
