共聚焦显微镜系统在蒿草切片样本宏观成像及微观成像中使用价值 被引量:1

Study on the Application Value of the Confocal Microscopy System in the Macro Imaging and Micro Imaging of Wormwood Slice Samples

摘要 目的研究探讨共聚焦显微镜系统宏观成像和微观成像在生物组织样本成像中的使用价值。方法使用共聚焦显微镜系统对蒿草切片样本进行宏观成像和微观成像,实验分为四组:A:蒿草切片样本放大1. 25倍成像; B:蒿草切片样本放大5倍成像; C:蒿草切片样本放大40倍局部视野成像; D:使用共聚焦显微镜系统视野拼图采图模式,4×4方格成像共16张,单张方格图片为放大160倍成像,再拼接成一张视野图。结果共聚焦显微镜系统不但可以获取1. 25倍和5倍蒿草切片样本宏观图像,显示蒿草切片样本整体平面结构,而且可以获取40倍蒿草切片样本局部视野图像,进一步可以获取放大160倍蒿草切片样本局部微观图像,并可对多张微观图进行拼接。结论共聚焦显微镜系统可对生物组织样本进行宏观成像和微观成像,与宏观成像比较,微观成像更有利于对生物组织样本的细微结构进行观察,结合视野拼图成像技术,具有较好的使用价值。 Objective To study the application value of macro imaging and micro imaging with confocal microscopy system in the fluorescence image of biological tissue sample. Methods The macro imaging and micro imaging of wormwood sample were performed with confocal microscopy system. The experiment was divided into four groups: group A, wormwood slice sample is magnified 1.25X;group B, wormwood slice sample is magnified 5X;group C, wormwood slice sample is magnified 40X local field imaging;group D, using the pattern of image stitching, 16 images are taken in 4 x 4 squares from local field wormwood slice sample, which are magnified 160X, and at micro levels are generated overview stitching image of local field wormwood slice sample. Results Confocal microscopy system not only can obtain 1.25X and 5X macro imaging which are revealed wormwood slice sample of the overall plane structure, but also can obtain 40 X local field imaging. Moreover, confocal microscopy system can obtain 160 X local field micro imaging and at micro levels generate overview stitching image. Conclusion Confocal microscopy system can obtain macro imaging and micro imaging of biological tissue sample. Compared with macro imaging, micro imaging can better observe the micro structure of biological tissue sample.
