摘要
目的:建立超高效液相色谱法同时测定小儿热速清颗粒中黄芩苷、黄芩素、葛根素、大豆苷、绿原酸及连翘苷6个成分的含量。方法:采用ZORBAX Eclipse Plus C_(18)色谱柱(2.1 mm×100 mm,1.8μm),流动相为乙腈-0.05%磷酸水溶液,梯度洗脱,流速为0.4 ml/min,检测波长为330 nm(0~5 min)、254 nm(5~9 min)、280 nm(9~18 min),柱温为40℃,进样量为1μl。结果:黄芩苷、黄芩素、葛根素、大豆苷、绿原酸及连翘苷的线性范围分别为8.4~84.0、1.1~11.0、8.6~86.0、2.1~21.0、2.0~20.0及2.0~20.0μg/ml,r≥0.999 0;平均加样回收率(n=9)分别为99.41%、101.56%、98.66%、101.61%、98.64%及101.71%;平均含量分别为17.44~19.82、0.37~0.66、0.78~0.97、3.04~4.16、0.51~0.98及0.39~0.42 mg/g。结论:该方法快速、准确,可用于小儿热速清颗粒的质量控制。
OBJECTIVE: To establish ultra performance liquid chromatography(UPLC) to determinate the contents of baicalin, baicalein, puerarin, daidzein, chlorogenic acid and forsythin in Xiaoer Resuqing granules. METHODS: ZORBAX Eclipse Plus C18 column(2.1 mm × 100 mm, 1.8 μm) was adopted, with mobile phase of acetonitrile-0.05% phosphoric acid aqueous solution, gradient elution, flow rate of 0.4 ml/min, detected wavelength of 330 nm(0-5 min), 254 nm(5-9 min), 280 nm(9-18 min), column temperature of 40 ℃, sample size of 1 μl. RESULTS: The linear ranges of baicalin, baicalein, puerarin, daidzin, chlorogenic acid and forsythin were respectively 8.4-84.0, 1.1-11.0, 8.6-86.0, 2.1-21.0, 2.0-20.0 and 2.0-20.0 μg/ml, r≥0.999 0;the average recovery rates(n=9) were respectively 99.41%, 101.56%, 98.66%, 101.61%, 98.64% and 101.71%;the average content were respectively 17.44-19.82, 0.37-0.66, 0.78-0.97, 3.04-4.16, 0.51-0.98 and 0.39-0.42 mg/g. CONCLUSIONS: This method is fast and accurate,which can be used for quality control of Xiaoer Resuqing granules.
