基于网络药理学的红花如意丸治疗妇科疾病作用机制研究 被引量:17

摘要 目的:基于网络药理学方法,初步探讨红花如意丸治疗妇科疾病的作用机制。方法:依托中药系统药理学数据库和分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)、中草药综合数据库(traditional Chinese medicine database@Taiwan,TDT)以及中国知网(CNKI),检索红花如意丸中主要药物所含的化学成分、作用靶点以及相关疾病,构建"化合物-靶点"网络、"靶点-疾病"网络、"药物-化合物-靶点-疾病"网络、蛋白质-蛋白质相互作用(protein-protein interaction,PPI)网络和"靶点-通路"网络,研究红花如意丸治疗妇科疾病的作用机制。结果:"化合物-靶点"网络中包含73个化合物和99个相应靶点,关键靶点涉及前列腺素内过氧化物合成酶2、前列腺素内过氧化物合成酶1和热休克蛋白90等;PPI网络中包含97个蛋白,关键蛋白涉及丝裂原活化蛋白激酶1(mitogen-activated protein kinase 1,MAPK1)、丝裂原活化蛋白激酶8(mitogen-activated protein kinase 8,MAPK8)和肿瘤蛋白p53(tumor protein p53,TP53)等;基因本体(gene ontology,GO)条目37个,其中生物过程相关的条目22个,分子功能相关的条目7个,细胞组成相关的条目8个;京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路14条,涉及神经活性配体-受体相互作用通路、恶性肿瘤的途径、雌激素信号通路和钙离子信号通路等。结论:本研究结果初步阐释了红花如意丸治疗妇科疾病的基本药理作用及其作用机制,为进一步深入揭示其作用机制奠定了良好的基础。 OBJECTIVE: To tentatively investigate the mechanism of Safflower Ruyi pills in treatment of gynecological diseases based on network pharmacology. METHODS: Traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP), traditional Chinese medicine database @Taiwan(TDT) and CNKI were retrieved for the chemical components, corresponding targets and diseases related to Safflower Ruyi pills. Compound-target network, target-disease network, TCM-compound-target-disease network, protein interaction(PPI) network,and target-pathway network were established to explore the mechanism of Safflower Ruyi pills in treatment of gynecological diseases. RESULTS: The compound-target network included 73 chemical components and 99 targets, the hub targets involved prostaglandin G/H synthase 2(PTGS2),prostaglandin G/H synthase 1(PTGS1) and heat shock protein HSP 90(HSP90), etc. The PPI network contained 97 proteins,in which the hub proteins involved mitogen-activated protein kinase 1(MAPK1), mitogen-activated protein kinase 8(MAKP8) and tumor protein p53(TP53), etc. Totally 37 GO terms were gene ontology(GO), among them, there were 22 items related to biological processes, 7 items related to molecular function, and 8 items related to cell composition. The Kyoto encyclopedia of genes and genomes(KEGG) pathway contained 14 pathways, including neuroactive ligand-receptor interaction, pathways in cancer, estrogen signaling pathways and calcium signaling pathways, etc. CONCLUSIONS: The results of this study preliminarily elucidate the basic pharmacological action and its mechanism of Safflower Ruyi pills in treatment of gynecological diseases, which lay a solid foundation for further studies on the mechanism of action.
