温针灸腰阳关、次穴对痛经模型大鼠子宫HIS及其H_1R表达影响 被引量:10

Effect of Warming Acupuncture on the Expression of HIS and H_1R in Rats Mode with Primary Dysmenorrhea

摘要 目的:观察温针灸腰阳关、次髎穴对痛经模型大鼠子宫内组织中组织胺(HIS)及其H1受体蛋白(H1R)水平的影响。方法:将72只雌性SD大鼠,随机分为造模组(60只)和空白组(12只),用缩宫素制备大鼠痛经模型,造模成功后,将60只痛经模型大鼠随机分为温针灸A组、温针灸B组、西药C组、西药D组、模型组。比较各组大鼠扭体反应的潜伏期和30 min内扭体次数,用酶联免疫吸附(ELISA)法、蛋白质免疫印迹(Wb)法检测各组大鼠子宫组织胺含量及其受体H1蛋白水平。结果:(1)空白对照组无扭体,模型组扭体次数较多,且有扭体潜伏期,两组比较有非常显著性意义(P<0.01)。与模型组相比,A组、B组、C组均能明显延长首次扭体潜伏期,且能减少30 min内扭体反应次数(P<0.01);而模型组与D组比较,扭体潜伏期、扭体次数差异无统计学意义(P>0.05)。A组、B组、C组与D组比较,D组首次扭体潜伏期最短,30 min内扭体次数最多(P<0.01)。A组、B组、C组扭体的潜伏期比较,差异无统计学意义(P>0.05);B组与C组均能减少30 min内扭体反应次数(P>0.05);但与B组、C组相比,A组30 min内扭体反应次数最少(P<0.05)。(2)与空白组比较,模型组、A组、B组、C组及D组HIS含量明显增多,H_1R水平显著降低(P<0.01)。与模型组、D组比较,A组、B组、C组HIS含量明显减少,H_1R表达明显升高(P<0.01)。A组、B组、C组的HIS、H1R水平比较,差异无统计学意义(P>0.05);但与B组相比,A组、C组HIS含量减少(P>0.05),H_1R表达上调(P>0.05);且与C组比较,A组HIS含量最少,H_1R表达最高。结论:温针灸腰阳关、次髎穴治疗原发性痛经与调节子宫HIS含量及H1R蛋白水平有关,即可下调痛经大鼠子宫HIS含量,提高H_1R蛋白水平。 Objective:To observe the effect of warm acupuncture and acupuncture on the level of histamine(HIS)and its H1 receptor protein(H1R)in the uterine tissue of the rat model of dysmenorrhea.Methods:72 female SD rats were randomly divided into group(60 rats)and blank group(12 rats). The rat model of dysmenorrhea was prepared by oxytocin. After the model was successful,60 rat model rats were randomly divided into warm moxibustion A group,warm moxibustion B group,Western medicine group,Western medicine D group and model group. The incubation period of writhing reaction and the number of torsional bodies in 30 min were compared. The content of histamine and the level of H1 protein in the uterus were detected by enzyme linked immunosorbent assay(ELISA)and protein immunoblotting(Wb).Results:There was no torsion in the blank control group. The number of twisting bodies in the model group was more than that in the control group,and there was a significant latency in the two groups(P<0.01).Compared with the model group,the A group,the B group and the C group could obviously prolong the incubation period of the first twisting body and reduce the number of twists in 30 min(P<0.01),but there was no statistical difference between the model group and the D group(P>0.05). In group A,group B and group C,compared with group D,the first period of writhing in D group was the shortest,and the number of twisting bodies in the 30 min group was the highest(P<0.01). There was no significant difference in the incubation period between group A,group B and group C(P>0.05),and in group B and C,the number of twists in 30 min was reduced(P>0.05),but in the B group and C group,the number of torriper reactions in A group 30 min was the least. Compared with the blank group,the HIS content in model group,A group,B group,C group and D group increased significantly,and H1R level decreased significantly(P<0.01).Compared with model group and D group,HIS content in A group,B group,C group and D group decreased significantly,H1R expression increased significantly(P<0.01). There was no significant difference in the level of HIS and H1R in group A,B group and C group(P>0.05),but compared with group B,HIS content in A group and C group decreased(P>0.05)and H1R expression was up(P>0.05). Conclusion:The treatment of primary dysmenorrhea and the regulation of the level of HIS and the level of H1R protein in the treatment of primary dysmenorrhea by warming acupuncture and acupuncture point can down regulate the content of HIS in the uterus of rats with dysmenorrhea and improve the level of H1R protein.
