摘要
为研究核桃果实油脂合成与积累的分子调控机理,以核桃果实种仁cDNA为模板克隆得到油脂合成关键转录因子JrWRI1基因,其cDNA序列全长为1 230 bp,编码409个氨基酸。通过对其结构特征、生物学特性进行分析,发现该基因编码蛋白属于AP2/ERF转录因子超级家族,具有特异的DNA结合序列,调节生物合成和代谢过程,定位于细胞核中,符合转录因子行使功能的特点。并构建了JrWRI1超表达重组载体,为进一步研究JrWRI1在核桃果实油脂合成与积累过程中的功能奠定基础,也为利用分子手段加快高油核桃新品种的选育提供参考。
In order to study the molecular regulation mechanism of lipids synthesis and accumulation in walnut fruits, a gene JrWRI1, the key transcription factor for lipids synthesis, was cloned from walnut seeds. The cDNA sequence of JrWRI1 was 1 230 bp encoding 409 amino acids. Through the analysis of structural and biological characteristics, it was found that the gene JrWRI1 belonged to the AP2/ERF transcription factor superfamily, which had specific DNA binding sequence, regulated the biosynthesis and metabolic process, and was located in the nucleus, so it was conformed to the characteristics of transcription factor performing functions. The over-expression recombinant vector of JrWRI1 was constructed, which laid a foundation for further study on the functions of JrWRI1 in the oil synthesis and accumulation in walnut seeds, and provided a reference for accelerating the selection and breeding of new varieties of high-oil walnut by molecular means.
