摘要
目的:探讨miR-146a在原发性肝癌组织中表达,以及上调原发性肝癌HepG2细胞中miR-146a表达水平对细胞增殖和侵袭的影响。方法:2015年3月至2018年3月,行肝癌手术治疗的原发性肝癌患者89例,利用实时荧光定量PCR技术检测原发性肝癌和癌旁组织中miR-146a表达,培养HepG2细胞并分为miR-146a模拟物组、阴性对照组和空白组,利用实时荧光定量PCR技术检测细胞中miR-146a表达,MTT法检测细胞增殖能力,Transwell法检测细胞迁移和侵袭能力。结果:原发性肝癌组织中miR-146a相对表达量为0.36±0.09,癌旁组织中则为1.04±0.11,差异有统计学意义(t=43.947,P=0.000);miR-146a表达量在中高分化、TNM分期Ⅲ~Ⅳ期、发生淋巴结转移及有门静脉癌栓的原发性肝癌组织中降低(P <0.05);miR-146a模拟物组细胞中miR-146a相对表达量高于阴性对照组和空白组(P <0.05);miR-146a模拟物组24、48、72和96h时细胞吸光度A值低于阴性对照组和空白组(P <0.05);miR-146a模拟物组迁移细胞数和侵袭细胞数低于阴性对照组和空白组(P <0.05)。结论:miR-146a在原发性肝癌组织中呈低表达,且与肿瘤发生及恶性化进展有关,特异性提高HepG2细胞中miR-146a表达水平可降低细胞增殖能力。
Objective:To investigate the expression of miR-146a in hepatocellular carcinoma tissues,and to explore the effect of up-regulating miR-146a expression level on cell proliferation and invasion in hepatocellular carcinoma HepG2 cells.Methods:A total of 89 cases of patients with hepatocellular carcinoma who underwent liver cancer surgery in our hospital were selected from March 2015 to March 2018.The expressions of miR-146a in hepatocellular carcinoma and adjacent normal tissues were detected by real-time fluorescent quantitative PCR.HepG2 cells were cultured and divided into miR-146a mimic group,negative control group and blank group.Real-time quantitative PCR was used to detect the expression of miR-146a in cells.MTT assay was used to detect cell proliferation,and Transwell assay was used to detect cell migration and invasion.Results:The relative expression level of miR-146a in hepatocellular carcinoma tissues was(0.36±0.09),while that in adjacent tissues was(1.04±0.11),the difference was statistically significant(t=43.947,P=0.000).The expression of miR-146a in hepatocellular carcinoma tissues was decreased in moderately high differentiation,TNM stageⅢ~Ⅳ,lymph node metastasis,and portal vein tumor thrombus.The relative expression level of miR-146a in the miR-146a mimic group was higher than that in the negative control group and the blank group(P<0.05).The cell absorbance A values at 24,48,72 and 96 h in the miR-146a mimic group were lower than those in the negative control group and the blank group(P<0.05).The number of migrating cells and the number of invasion cells in the miR-146a mimic group were lower than those in the negative control group and the blank group(P<0.05).Conclusion:miR-146a was lowly expressed in hepatocellular carcinoma tissues and was associated with tumorigenesis and malignant progression.Specific increased the expression of miR-146a in HepG2 cells could reduce cell proliferation and inhibit cell migration and invasion.
