摘要
目的观察糖痹康对糖尿病大鼠背根节神经组织氧化应激、细胞凋亡的影响,并探讨其机制。方法选取SD雄性大鼠60只,随机分为正常对照组、模型组、糖痹康低剂量组、糖痹康中剂量组、糖痹康高剂量组及弥可保组各10只。模型组、糖痹康低剂量组、糖痹康中剂量组、糖痹康高剂量组及弥可保组采用腹腔注射链脲佐菌素法制备糖尿病模型,正常对照组腹腔注射等量注射枸橼酸盐缓冲液。8周后,将各组大鼠麻醉,取出背根神经节,检测背根神经节组织活性氧簇(ROS)水平,采用TUNEL法测算神经节组织细胞凋亡率,采用Western blotting法检测凋亡相关蛋白B细胞淋巴瘤/白血病-2(Bcl-2)、活化的含半胱氨酸的天冬氨酸蛋白水解酶3(Cleaved Caspase3)以及p38丝裂原活性蛋白激酶(p38 MAPK)信号通路相关蛋白p38、p-p38的表达。结果 (1)模型组ROS水平高于正常对照组,糖痹康低、中、高剂量组及弥可保组ROS水平均低于模型组(P均<0. 05)。(2)模型组细胞凋亡率高于正常对照组,糖痹康低、中、高剂量组及弥可保组细胞凋亡率均低于模型组(P均<0. 05)。(3)模型组Bcl-2蛋白表达低于正常对照组,Cleaved Caspase3蛋白表达高于正常对照组(P均<0. 05);与模型组比较,糖痹康低、中、高剂量组及弥可保组Bcl-2蛋白表达均升高,Cleaved Caspase3蛋白表达均降低(P均<0. 05)。(4)模型组p38、p-p38蛋白表达均高于正常对照组,糖痹康低、中、高剂量组及弥可保组p38、p-p38蛋白表达均低于模型组(P均<0. 05)。结论糖痹康可降低糖尿病大鼠背根神经节组织ROS水平、抑制细胞凋亡,其机制可能与抑制p38 MAPK信号通路有关。
Objective To investigate the effect of Tangbikang on apoptosis and oxidative stress in diabetic peripheral neuropathy(DPN)rats.Methods Sixty SD male rats were selected,and were randomly divided into the normal control group,model group,low-dose,medium-dose,high-dose Tangbikang groups,and methycobal group,with10in each.Diabetic model was established by intraperitoneal injection of streptozotocin(STZ)in the model group,low-dose,medium-dose,high-dose Tangbikang groups and methycobal group,while the normal control group,the rats were given intraperitoneal injection of citrate buffer.Eight weeks later,rats in each group were anesthetized,dorsal root ganglion was removed,ROS level in dorsal root ganglion nerve tissues was measured,apoptotic rate of nerve tissues was measured by TUNEL method,and Bcl-2,Cleaved Caspase3,p38,p-p38 protein expression levels were detected by Western blotting.Results①ROS level in the model group was significantly higher than those in the normal group,low-dose,medium-dose,high-dose Tangbikang groups,and methycobal group(all P<0.05).②The apoptosis rate in the model group was significantly higher than those of the normal group,low-dose,medium-dose,high-dose Tangbikang groups,and methycobal group(all P<0.05).③Bcl-2 protein expression was significantly lower and Cleaved Caspase 3 protein expression was significantly higher in the model group than in the normal group;compared with the model group,Bcl-2 protein expression in low-dose,medium-dose,high-dose Tangbikang groups and methycobal group was significantly higher,and Cleaved Caspase 3 protein expression was significantly lower(both P<0.05).④p38 and p-p38 protein expression levels in the model group were significantly higher than those of the normal group;p38 and p-p38 protein expression levels in the low-dose,medium-dose,high-dose Tangbikang groups and methycobal group were significantly lower(both P<0.05).Conclusion Tangbikang can reduce ROS level and inhibit apoptosis of dorsal root ganglion nerve tissues in diabetic rats by inhibiting the p38 MAPK signaling pathway.
