大孔树脂分离纯化罗勒叶总黄酮及抗氧化活性研究 被引量:11

摘要 以黄酮的吸附率、解吸率和回收率作为主要评价指标,对罗勒叶总黄酮纯化工艺进行研究。结果表明,在8种大孔树脂中选取X-5型用于纯化,最佳纯化工艺参数为:黄酮浓度为2.56 mg/mL时,56 mL pH值为4的上样液以2 BV/h的流速进行上样。待吸附饱和后,64 mL pH值为6的70%的乙醇溶液以2 BV/h的流速洗脱。工艺纯化后的纯度提高至69.36%,且对1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl radical,DPPH)和2'-联氨-双-3-乙基苯并噻唑啉-6-磺酸[2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid,ABTS)自由基半数清除率IC50值分别为0.215 mg/m L和0.544 mg/mL,明显高于粗黄酮的1.06 mg/mL和1.717 mg/mL。 The flavonoid adsorption rate,desorption rate and recovery rate were taken as the main evaluation indexes to study the purification process of total flavonoids in Ocimum basilicum L..The results showed that X-5 was selected for purification in 8 kinds of macroporous resins.We showed that the optimal purification conditions include flavonoids concentration of sample(2.56 mg/mL),pH value(4),the flow rate(2 BV/h),and the maximum sample volume(56 mL).After complete adsorption,the macroporous resin column was washed with 64 mL 70%alcohol and pH value(6),a flow rate of 2 BV/h.The purity after purification increased to 69.36%,and the IC50 values of 1,1-diphenyl-2-picrylhydrazyl radical(DPPH)and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid(ABTS)free radicals were 0.215 mg/mL and 0.544 mg/mL respectively,which were significantly higher than 1.06 mg/mL and 1.717 mg/mL for crude total flavonoids.
