低温体外循环术后犬心肌细胞凋亡及Fas/FasL系统的调节作用 被引量:4

摘要 目的 观察低温体外循环 (CPB)术后心肌细胞凋亡及Fas/FasL系统和氧化应激的调节作用。方法  1 0条成年家犬用于模拟低温CPB手术。实验动物随机分为对照手术组和CPB组 (n分别 =5)。分别在CPB术前、术中、术后 1和 3h留取左心室 (LV)标本。用原位末端脱氧核糖核苷酸转移酶标记 (TUNEL)技术检测心肌凋亡细胞。分别用免疫组织化学和流式细胞仪(FACS)技术定性和定量测定心肌细胞Fas和FasL蛋白表达。用硫代硫酸钠法测定LV脂质过氧化物含量。结果 CPB术后 ,CPB组心肌细胞凋亡数较术前、术中和对照手术组均明显增高 (P值均 <0 .0 1 )。免疫组织化学结果显示Fas和FasL是心肌细胞胞浆的固有组成成分。FACS结果证实CPB术后左心室Fas和FasL蛋白表达明显上调 (P值均 <0 .0 1 )。LV脂质过氧化物含量也明显增高 (P值均 <0 .0 1 )。结论 低温CPB可以诱导 (犬 )心肌细胞凋亡。 Objective To investigate the myocardial cell apoptosis in dog hearts following hypothermic cardiopulmonary bypass (CPB) and modulation of Fas/FasL pathways and role of myocardial oxidative stress in this setting.Methods Ten adult dogs were randomized into sham operated and CPB groups. Samples of left ventricle (LV) were obtained before CPB,during CPB,1 h and 3 h after CPB.In situ TUNEL was used to detect apoptotic myocardial cell. Immunohistochemistry and flow cytometry were employed to detect the expression of Fas and FasL.TBARS method was applied to measure the content of lipid peroxide.Results After CPB,the number of apoptotic myocardial cells in LV was significantly increased as compared with that of before and during CPB and that in the sham operated group (P<0.01,respectively). Immunohistochemistry demonstrated that Fas and FasL proteins were constitutively present on the sarcolemma of LV myocytes. FACS results showed that the expression of Fas and FasL was significantly upregulated after CPB (P<0.01,respectively). The content of lipid peroxide in LV was also significantly increased (P<0.01,respectively).Conclusion Hypothermic CPB induces myocardial cell apoptosis in dog hearts and the CPB induced myocardial cell apoptosis was associated with upregulation of expression of Fas and FasL and increase of oxidative stress in the myocardium.
