摘要
目的探讨川芎嗪对大鼠肝星状细胞系HSC-T6自噬的影响。方法应用蛋白免疫印迹法检测不同浓度的川芎嗪对大鼠肝星状细胞系HSC-T6自噬蛋白表达的影响及相关自噬分子蛋白的表达情况,使用MDC染色检测川芎嗪对肝星状细胞系HSC-T6自噬溶酶体的形态结构影响。结果 25 ng/ml浓度的川芎嗪对大鼠肝星状细胞系HSC-T6的作用效果最佳。川芎嗪组细胞球数目少于对照组,肝星状细胞系HSC-T6 ATG5表达水平低于对照组,beclin1及LC3Ⅱ表达水平高于对照组(P<0.05)。结论川芎嗪可以影响大鼠肝星状细胞系HSC-T6的自噬溶酶体,减弱自噬的发生。
Objective To investigate effect of Ligustrazine on autophagy of hepatic stellate cell line HSC-T6 in rats.Methods Western blotting was used to detect effect of different concentrations of Ligustrazine on autophagy protein expression and related autophagy molecule protein expression of hepatic stellate cell line HSC-T6 in rats.Effect of Ligustrazine on morphological structure of autophagolysosome of hepatic stellate cell line HSC-T6 in rats was detected by MDC staining.Results The effect of Ligustrazine on hepatic stellate cell line HSC-T6 in rats was the best when Ligustrazine concentration was 25 ng/ml.In Ligustrazine group,number of cell ball was significantly decreased,and expression of hepatic stellate cell line HSC-T6 ATG5 was significantly lower,while beclin1 and LC3Ⅱexpressions were significantly higher than those in control group(P<0.05).Conclusion Ligustrazine may affect autophagolysosome of hepatic stellate cell line HSC-T6 in rats to decrease incidence rate of autophagy.
