摘要
目的建立铁皮石斛干、鲜品细胞破壁率的检测方法,并考察铁皮石斛不同干品粉末和鲜品榨汁破壁率与溶出量的关系。方法采用细胞计数法,以薄壁细胞为显微特征指标,测定铁皮石斛5种不同粉末(粗粉、50目粉、最细粉、极细粉、微粉)和鲜品榨汁(以下简称鲜汁)的破壁率;以多糖为指标,考察上述粉末和鲜汁破壁率与其多糖溶出量的关系。结果在2 h内,铁皮石斛粉末和鲜汁的破壁率与其溶出量有较强的正相关性,相同质量铁皮石斛中多糖的溶出量随破壁率的升高而提高,微粉和鲜汁的破壁率达到100%时,多糖溶出量按干燥品计分别为45.25%和54.10%,鲜品的多糖溶出量高于干品。结论随着破壁率的提高,铁皮石斛多糖的溶出速度加快、溶出量增加,能有效地节省提取时间,提示鲜药多糖的利用率明显高于干品。
Objective To establish a method for the determination of cell-broken rate in fresh and dried Dendrobium officinale,and investigate the relationship between the fragmentation efficiency and the rate of dissolution of dried powder and fresh product.Methods Five kinds of different powders(coarse powder,50 mesh powder,the most fine powder,very fine powder and micro powder)and fresh product(hereinafter referred to as fresh juice)were prepared using cell counting method and parenchyma cells as evaluation indexes of the broken rate.Polysaccharide was employed as an indicator to investigate the relationship between broken rate and the amount of polysaccharide dissolution.Results In 2 hours,the broken rate of D.officinale powder and fresh juice has a strong correlation with the dissolution rate.The dissolution rate of polysaccharides increased with the wall-broken rate,and the dissolution rates of polysaccharides were 45.25%and 54.10%in drying meter when the rate of breakage of powder and juice was 100%.The amount of fresh product was higher than dry products.Conclusion The dissolution speed of polysaccharide from D.officinale increased significantly with the broken rate,which could effectively save the extracting time.It is suggested that the utilization rate of polysaccharides of fresh products is obviously higher than that of dry goods.
