基于网络药理学预测的芪参颗粒干预过量异丙肾上腺素诱导小鼠心力衰竭的机制研究 被引量:7

Study on Mechanism of Qishen Granules on Heart Failure Induced by Excess Isoproterenol in Mice Based on Network Pharmacological Prediction

摘要 目的探讨芪参颗粒干预过量异丙肾上腺素致小鼠心力衰竭的效果,并基于网络药理学的方法预测其作用机制。方法制备过量盐酸异丙肾上腺素诱导的心力衰竭小鼠模型,随机分为模型组、空白组、芪参颗粒组、福辛普利组。通过超声、病理和心脏指数分析各组小鼠心功能,并采用生物信息学分析工具(BATMAN-TCM)预测靶点。结果超声方面,芪参颗粒可显著降低心力衰竭小鼠左室收缩末内径(P<0.05),显著提高射血分数(P<0.05)与短轴缩短率(P<0.05)。形态学中,芪参颗粒可有效降低心力衰竭小鼠心脏指数。病理显示芪参颗粒组心肌组织的病变程度显著减轻。网络药理学显示芪参颗粒治疗过量异丙肾上腺素诱导的心力衰竭作用靶点可能与血管紧张素转化酶(ACE)、α2A型肾上腺素受体(ADRA2A)、α-2型肾上腺素受体B亚型(ADRA2B)等有关。结论芪参颗粒能显著改善过量异丙肾上腺素诱导的小鼠心力衰竭,其机制可能与血管紧张素Ⅱ(AngⅡ)降低,抑制肾素血管紧张素系统(RAS)活性、调节交感神经系统、抑制肿瘤坏死因子(TNF)等炎症介质合成等有关。 ObjectiveTo study the effect of Qishen granules(QSG)on heart failure and its related targets by network pharmacological predicaiton.MethodsHeart failure model was induced by over-dose isoproterenol in C57 mice.Then the mice with heart failure were randomly divided into model group,QSG group,and fosinopril group,while a sham group was included.After 14 days of treatment,cardiac index,echocardiography,and hematoxylin eosin staining of cardiac tissue were detected,and the targets of QSG was predicted by Bioinformatics Analysis Tool for Molecularmech ANism of Traditional Chinese Medicine(BATMAN-TCM).ResultsComparing with model group,left ventricular internal systolic diameter was significantly reduced in QSG group,while the ejection fraction and fractional shortening were dramatically improved(P<0.05).Pathology also revealed that the lesions in myocardium was significantly reduced in QSG group.Network pharmacological predication showed that multiple targets were related to the effect of QSG,including angiotensin-converting enzyme(ACE),DRA2A,and adrenoceptor alpha 2A,ConclusionQSG can significantly improve the heart function with in mice heart failure induced by over dose isoproterenol.These effects may be related to the reduction of angiotensinⅡ,the inhibition of rennin-angiotensin system(RAS)activity,the regulation of sympathetic nervous system,and catecholamine system,the inhibition of TNF and other inflammatory mediators,etc.
