小麦叶内锈菌细胞核荧光染色方法 被引量:2

A Method of Flourescent Staining for Nuclei of Rust Hyphae in wheat Leaves

摘要 本文对小麦叶内锈菌菌丝细胞核的荧光染色方法进行了研究。受锈菌侵染的小麦叶片经0.25%纤维素酶和0.12%Driselase混合液培养2~3h,锈菌菌落从组织中游离出。菌落经荧光染料DAPI(5μg/mL)和Calcoflour white(0.001%)分别染色1~3min后,菌丝的细胞核、壁、隔膜在荧光显微镜下易于观察辩认。 This paper describes a method of Flourescent staining for nuclei of rust hyphae in wheat leaves.Rust colonies were freed from leaf tissue after rust-infected wheat leaves had been incubated for 2 to 3 h.with the mixture of 0.25% cellulase and 0.12% driselase.These colonies were then stained for 1 to 3 min.with DAPI (5ug/mL) and Calcoflour white (0.001%) respectively, and the nuclei, Cell wall and septa of rust hyphae could be easily detected under flourescent microscope.
