摘要
PGD_2的合成子(9)的合成是以2-甲基环戊1,3-二酮(1)为原料,经烯醇醚化、烯丙基化、酸去保护、羧基的锂铝氢还原、臭氧化、氧化、甲酰化获得。
The isobutyl enol ether (2) of 2-methyl cyclopentandione-1, 3 (1)used as starting
material was alkylated with allylbromide to form 3. Hydrolysis of 3 afforded the dione
4, which on reduction with LiAlH_4 gave oily diols 5. Ozonolysis of 5 inthe presence
of Na_2CO_3 in methanol and dichloromethane at -78℃ produced the mixture of
hemiacetals 6, which were oxidized to the compound 7 by Jones reagent. To introduce
an aldehyde group next to the ketone position, 7 was reacted with ethyl formate
followed by treatment with p-TsOH to give 9 which is a synthon for synthesis of 10-
methyl-PGD_2.
