摘要
目的 :探讨我国 4省三带喙库蚊的遗传多态性。方法 :分别用 8种随机引物对采自我国广东、福建、海南和贵州的 12只三带喙库蚊的基因组DNA进行聚合酶链反应 (PCR)扩增 ,1.5 %琼脂糖凝胶电泳检测结果。用PHYLIP软件包中的NEIGHBOR程序对随机扩增多态性DNA数据进行聚类分析 ,然后用软件TREEVIEW绘制出系统发育树。结果 :8种随机引物共扩增出RAPD片段 2 70个 ,RAPD谱带均为单型。系统发育树显示 ,海南的 2只三带喙库蚊聚为一枝 ,而其余标本中同一省份的标本均未能聚为独立的分枝。结论 :三带喙库蚊个体间的遗传变异较大 ;不同省份三带喙库蚊的变异具有交叉。
Objective:To study the genetic polymorphism of Culex tritaeniorhynchus from four provinces in China. Methods:The genomic DNA of 12 Cx. tritaeniorhynchus samples collected from Guangdong, Fujian, Hainan and Guizhou were amplified separately with 8 random primers The PCR results were detected with 1.5% agarose electrophoresis. The clustering analysis of the randomly amplified polymorphic DNA data was processed with NEIGHBOR program of PHYLIP, and the phylogenetic tree then was drawn with software TREEVIEW. Results: 270 RAPD fragments were obtained and the RAPD strap style of each individual was monotype. The phylogenetic tree showed that two samples from Qiongzhong clustered into a branch But among some other samples, which came from same province,did not cluster into independent branchs. Conclusion:The genetic varieties among Cx. tritaeniorhynchus individuals are relatively rich. At the same time, there are cross in genetic varieties among geographic populations from different provinces.
