摘要
目的 了解hERR1在乳腺癌及癌旁组织中的表达情况 ,以及它对芳香族化酶基因表达的调节作用。方法 收集乳腺癌患者的临床标本 ,应用RT PCR法 ,研究癌组织及相应的癌旁组织中hERR1的表达情况 ;采用瞬时转染和CAT活性分析的方法 ,研究hERR1对芳香族化酶基因表达的调节。结果 hERR1在癌组织中的表达明显高于相应的癌旁组织 (16 2 6例 ) ;hERR1以剂量依赖方式促进芳香族化酶启动子Ⅰ 3活性。结论 hERR1在体内可能起到促进乳腺癌生长的作用。
Objective To study the expression of hERR1 in breast cancer tissues and para carcinoma tissues, and its effects on aromatase gene expression Methods RT PCR was applied to analyze the expression of hERR1 in breast cancer tissue and para carcinoma tissue Effects of hERR1 on aromatase gene expression were studied with transient transfection and CAT activity assay Results hERR1 expressed in both normal and malignant breast tissues, with higher levels in carcinoma tissues than in corresponding peri carcinoma tissues in the most of samples examined The transient transfection assays showed that hERR1 increased CAT transcription under the control of promoter 1 3 of the human aromatase gene in a dose dependent manner Conclusion hERR1 might increase the growth rate of breast cancer in vitro
