二甲基亚硝胺诱发大鼠肝纤维化过程中α平滑肌肌动蛋白与门脉压力的变化 被引量:17

Changes of α-smooth Muscle Actin and Portal Vein Pressure during Dimethylnitrosamine-induced Hepatic Fibrosis in Rats

摘要 目的 :观察肝纤维化形成过程中 α平滑肌肌动蛋白 (α- SMA)与门脉压力变化及两者的关系。方法 :采用二甲基亚硝胺 (DMN)诱发大鼠肝纤维化模型 ,共 4周。分别于造模开始后 2 d,1、2、3、4、5、6、8、12及 2 4周末观察大鼠肝纤维化程度、α- SMA表达与门脉压力。结果 :造模过程中肝纤维化程度逐渐加重 ,至第 4周末最为明显 ,其后逐渐减轻 ;α- SMA表达与肝纤维化程度的变化相一致 ,两者呈正相关 ;门脉压力的变化与纤维化程度及 α- SMA表达均呈正相关。结论 :肝星状细胞 (HSC)的活化在肝纤维化形成中起着重要作用 ,并对门脉压力起调节作用。 objective: To investigate the changes of α smooth muscle actin (α SMA) and portal vein pressure and their correlation during dimethylnitrosamine (DMN) induced hepatic fibrosis. Methods: The fibrotic model was induced by DMN for 4 weeks. At 2 day, 1 week, 2 week, 3 week, 5 week, 6 week, 12 week and 24 week after DMN intoxication, the fibrotic degree, α SMA expression and portal vein pressure were observed respectively. Results: The fibrotic degrees were gradually increased during the induction of the model, reached its peak at 4 week, and then gradually decreased.α SMA expression was positively correlated with hepatic fibrosis. The portal vein pressure was positively correlated with α SMA expression and hepatic fibrosis. Conclusion: Activation of hepatic stellate cells play an important role in hepatic fibrogenesis and regulate the portal pressure.
