摘要
目的 :观察化学致畸、致癌物乙酰基亚硝基脲 (ethylnitrosourea ,ENU)对体外培养牙胚发育、分化的影响。方法 :通过体外牙胚器官培养模型观察培养液中不同浓度的ENU对 1 7天胎龄的小鼠下颌第一磨牙牙胚形态发育的影响。结果 :牙胚培养 6天后 ,在含 1 0 %小牛血清的BGJb培养液中 ,牙胚的内釉上皮细胞分化为成釉细胞并分泌牙釉质基质 ,而其下方的牙乳头细胞分化为成牙本质细胞并分泌前期牙本质。当BGJb培养液中含 0 0 2 5 %ENU时 ,牙胚成釉器的星网层发生液化、囊性变。当BGJb培养液中含 0 0 5 %ENU时 ,牙胚发育受到抑制 ,其完整性被破坏 ,部分细胞溶解坏死。结论 :一定量的ENU可使发育中的牙胚发生囊肿样变 。
Objective:To investigate histologically the effects of carcinogenic ethylnitrosourea on the mouse tooth germs in organ culture.Methods:The mouse tooth germs were divided into four groups and cultured in Trowell type organ culture system.The effects of different ENU concentrations in BGJb medium on the germs of mandibular first molars in embryonic day 17 mice were examined.Results:The inner enamel epithelium of the tooth germ after 6 days of culture in BGJb medium with 10% fetal bovine serum had differentiated into ameloblasts which formed the enamel matrix,while the dental papilla cells under the inner enamel epithelium had differentiated odontoblasts which secreted predentin.The stellate reticulum of the tooth germ after 6 days of culture in BGJb medium containing 0 025% ENU and 10% fetal bovine serum was liquescent and formed cysts.The development of the tooth germ was prohibited and parts of the tooth germ showed necrosis after 6 days of culture in BGJb medium containing 0 05% ENU and 10% fetal bovine serum.Conclusion:The results suggest that 0 025% ENU in BGJb medium make the tooth germ cystic and 0 05% ENU in BGJb medium has toxic effects on the tooth germs in vitro.
