摘要
本文叙述了激光扫描共聚焦显微镜的光学原理,以花粉颗粒为例,利用激光扫描共聚焦显微镜对生物组织进行连续“光学切片”断层扫描,并重建其三维结构。结果显示重建后的图像为高分辨率的三维原色图像,可沿着X、Y、Z轴旋转或其它任意角度显示。
The optical principles of laser scanning confocal microscopy (LSCM) were described in this paper. As an example, a sample of pollen particles stained with FITC was analyzed by using the “optial sectioning” CT function of the LSCM. The 3D image of the biological structure of the pollen particle was then reconstructed with the data processing system. The reconstructed 3D image of the pollen particle was of a primary color image with high resolving power, and could be rotated and observed from any angle in the 3D space.
