3Dimensional reconstruction of pollen particles with laser scanning confocal microscopy

摘要 本文叙述了激光扫描共聚焦显微镜的光学原理,以花粉颗粒为例,利用激光扫描共聚焦显微镜对生物组织进行连续“光学切片”断层扫描,并重建其三维结构。结果显示重建后的图像为高分辨率的三维原色图像,可沿着X、Y、Z轴旋转或其它任意角度显示。 The optical principles of laser scanning confocal microscopy (LSCM) were described in this paper. As an example, a sample of pollen particles stained with FITC was analyzed by using the “optial sectioning” CT function of the LSCM. The 3D image of the biological structure of the pollen particle was then reconstructed with the data processing system. The reconstructed 3D image of the pollen particle was of a primary color image with high resolving power, and could be rotated and observed from any angle in the 3D space.
