褪黑素对大鼠局灶性脑缺血后梗塞体积和神经元形态变化的影响

Effects of MT on the Infarct Sizes and Neuronal Morphologic Change after Focal Cerebral Ischemia in Rats

摘要 目的探讨褪黑素 (MT)对大鼠局灶性脑缺血是否具有保护作用及其作用机制。方法用线栓法建立大鼠大脑中动脉栓塞 (MCAO)模型 ,大鼠随机分为缺血组 ,MT组 ( 1 0mg/kg) ) ,尼莫地平组 ( 0 .2mg/kg)。对术后 3h、6h及 2 4h三个时间点神经功能的缺陷评分后 ,处死动物 ,测定脑梗塞体积 ;2 4小时后作HE染色 ,观察神经元的病理形态学改变。结果MT可明显改善神经缺陷症状 ,缩小梗死体积 ,减轻脑组织的病理性损伤。结论MT对大鼠局灶性脑缺血有一定的保护作用 ,其机制可能与MT缩小梗死体积。 Objective To explore the protective effects and mechanism of melatonin(MT) on permanent focal cerebral ischemia.Methods The models of unilateral middle cerebral artery occlusion(MCAO) were established in rats,and randomly devided into MT(10mg/kg)group,Nimodipin(Nim,0.2mg/kg)group and control(ischemia)group.Brain damage was appreciated according to the neurological score,the infarct sizes and the histotogical changes induced by focal cerebral ischemia.Results Resembling to the Nim group,in MT group the neurological deficits were significantly attenuated after ischemia.Compared with ischemic group,the infarct volumes were obviously reduced and the histological changes were significantly diminished in MT group as well as Nim group.Conclusion This experiment suggests that MT may protect the brain from focal ischemia.
