一氧化氮合酶在急性肝功能衰竭大鼠主要器官中表达的变化

摘要 目的 :从分子和蛋白水平揭示在急性肝功能衰竭 (AL F)时不同一氧化氮合酶 (NOS)诱导产生的一氧化氮 (NO)在肝、肺、肾脏和肠组织中表达的变化。方法 :雄性 Wistar大鼠 6 0只 ,随机分为 6组 :假手术 (SO)组、AL F 6 h组、AL F 12 h组、L - Arg组、L - NAME组、L - Arg+L - NAME组 ,每组 10只 ,L - Arg用量 30 0 m g/kg,L - NAME用量 30 mg/kg;用原位杂交和免疫组化方法检测肝、肺、肾组织 e NOS m RNA、i NOS m RNA表达和小肠、大肠 e NOS和 i NOS表达。结果 :AL F 6 h组肺 e NOSm RNA表达明显增加 ,肝、肺、肾 i NOS m RNA表达亦显著增加 ,而 AL F 12 h组则明显降低 (P<0 .0 5 )。应用 L - Arg在 AL F 6h组肝、肾 i NOS m RNA表达明显降低 ,而应用 L - NAME和 (或 )应用 L - Arg在 AL F 6 h组肺 e NOS m RNA及肝、肺、肾 i NOSm RNA表达均显著降低 (P<0 .0 5 ) ;AL F 6 h组小肠、大肠黏膜 i NOS表达显著增加 ,而 e NOS表达在 6 h、12 h组明显降低 ,尤其在 12 h组降低更明显 (P<0 .0 5 ) ;应用 L - Arg和 (或 )应用 L - NAME时 ,小肠、大肠黏膜 e NOS、i NOS表达均明显降低(P<0 .0 5 ) ;应用 L- NAME大肠黏膜 e NOS、i NOS表达均明显降低 (P<0 .0 5 )。结论 :AL F早期肺、肾组织 i NOS m Objective:To observe the expression changes of nitric oxide synthase(NOS) in the liver,the lung,the kidney and the intestine of rat during acute liver failure(ALF).Methods:Wistar male rats(n=60) were randomly divided into 6 groups:SO group,ALF 6 h group,ALF 12 h group,L-Arg (300 mg/kg) group,L-NAME (30 mg/kg) group,L-Arg+L-NAME group, each group had 10 rats.The expression of eNOS mRNA and iNOS mRNA in the liver,lung and kidney was determined with in situ hybridization,and in the small and large intestines was determined with immunohistochemical method.Results:The expression of eNOS mRNA in the lung and iNOS mRNA in the liver,lung and kidney increased significantly 6 h after operation,but the expression of eNOS mRNA and iNOS mRNA in the liver,lung and kidney reduced 12 h after operation(P<0.05); The expression of iNOS mRNA in the liver and kidney reduced significantly with the reagents of L-Arg 6 h after operation,but the expression of eNOS mRNA in the lung and iNOS mRNA in the liver,lung and kidney reduced notably with the reagents of L-NAME or/and L-Arg 6 h after operation(P<0.05);Expression of iNOS in the small and large intestinal mucosa increased significantly 6 h after operation,but the expression of eNOS reduced 6 h,12 h,especially 12 h after operation(P<0.05);The expression of eNOS and iNOS in the small and large intestinal mucosa reduced significantly with the reagents L-Arg or/and L-NAME(P<0.05); The expression of eNOS and iNOS in the large intestine reduced notably because of NOS inhibition (P<0.05).Conclusion:The expression of iNOS mRNA in the lung and the kidney increases significantly at the early stage of ALF,the expression of eNOS in the small and large intestine decreases,but the expression of iNOS in the small and large intestine increases at the early stage of ALF,NO precursor can moderate the expression of iNOS mRNA in the liver,lung and kidney;The expression of iNOS in the small and large intestine reduces significantly with the reagents of L-Arg or/and L-NAME, moderating the damage of intestine mucosa, liver, lung and kidney.
