人原发性肝癌P53基因缺失和突变 被引量:8

摘要 在多种人体肿瘤组织中P53基因有缺失,P53基因作为一个抗癌基因,能在体外抑制肿瘤细胞的生长。肿瘤组织中P53基因缺失或发生突变,丧失了抗癌基因的活性,某些突变体还具有协同转化活性。本文分析了12例肝癌及其相应癌旁正常组织P53基因的存在状态,发现3例(25%)P53基因有缺失,3例P53基因有突变。这一结果提示P53基因为人肝癌抗癌基因,它的缺失或突变与肝癌的发生或演进有关。 Deletions and mutations of P53 gene occurred frequently in diverse human tumortypes, including human hepatocellular carcinoma. The tumorous and corresponding non-tumorous liver tissue of 12 patients from Qidong County were analyzed. 2 out of the 12samples showed loss of heterozygosity at 17p alleles, while the other one showed chro-mosome rearrangement in this region.No mutation in exon 7were detected in the aforementioned 3 samples by direct sequencing. Mutations between codon 249 and 250 of P53gene were observed in the tumorous tissue of another 3 samples. The sequence analysisof the rest tumorous and non-tumorous tissues is just under way
