光敏核不育杂交水稻Ks—9/直龙的种子纯度鉴定 被引量:4

摘要 种子纯度鉴定对杂交水稻增产关系十分密切。本文利用等电点聚焦电泳测同工酶法对光敏核不育杂交水稻Ks—9/直龙进行了种子纯度的鉴定。实验结果表明,早期萌发的ks—9/直龙其胚芽、胚根和胚乳的酯酶同工酶谱有明显的差异。其中,胚芽同工酶的种类和含量最多,能真实地显现出该品种的酶谱特征。ks—9/直龙的酶谱类型属单亲本型,即ks—9/直龙的酯酶同工酶谱与其父本直龙同型。室温下贮藏一年的ks—9/直龙种子在萌发初期,其酶的含量明显下降,且有些酶带消失,但还能体现出该品种的酶谱特征,表明酯酶同工酶较稳定,是种子纯度鉴定的好指标。 Purity of seeds is very important for HPGMR (photoperiodicsensitive Genie Male—sterile Rice) production. The esterase isozymes of seeds were determined by electrophoresis of isoelectric focusing. The number of isozymes and the activity of esterase in plumule were the largest and represented the characteristics of cultivars or strains. It was suggested that use of plumule as material for the electrophoresis to identify the cultivars will be more accurately. The isozyme pattern of KS-9/Zhi-Long was single-parent type corresponding to its father———Zhi-long cultivars. When seeds of KS-9/Zhi-long stored in room temperature for one year, viability and vigour decreased to a very low level. However, the isozyme patterns of esterase can still be maintained. Owing to the stability of the isozymes of esterase, the isozyme pattern can be used as an index in identification of cultivars or strains.
