摘要
目的 建立测定人尿中金雀异黄素和黄豆苷原含量的高效液相色谱方法。方法 采用了高效液相色谱法。条件为InertsilODS 34.6mm× 2 5 0mm ,5 μm色谱柱 ;流动相为甲醇∶水∶冰醋酸 (70∶30∶0 .2 ) ;检测波长2 6 0nm ;流速为 0 .7mL/min。结果 金雀异黄素的工作曲线范围 0 .0 2 84~ 1.4 2 μg/mL ,回归方程为 :ρ=97.18A +19.35 ;黄豆苷原的工作曲线范围 0 .0 4 32~ 2 .16 μg/mL ,回归方程为 :ρ =87.5 3A +2 6 .0 3。 结论 此方法提取简单 。
Purpose. To establish HPLC method to determine genistein and daidzein in human urine. Methods. HPLC method was used with Inertsil ODS-3 4.6 mm × 250 mm, 5 μm, the mobile phase was methanol: water: glacial acetic acid (70:30:0.2). Detection wavelength was 260 nm, flow rate 0.7 mL/min. Results. The linear range was 0.028 4 ∼ 1.42 μg/mL, the standard curve equation was ρ = 97.18A + 19.35 for genistein; and 0.043 2 ∼ 2.16 μg/mL, ρ = 87.53A + 26.03 for daidzein. Conclusions. The extraction is simple, and the method is accurate, rapid and reproducible.
