摘要
目的 研究成纤维细胞在糖尿病心肌纤维化病变中的作用。方法 用链脲佐菌素制备 10只糖尿病大鼠模型 ,饲养 12周后行大鼠心肌成纤维细胞的原代培养 ,计数细胞观察其成长 ,并分别采用RT PCR及westernblot检测α1(Ⅰ )前胶原的mRNA、蛋白表达。结果 糖尿病大鼠心肌成纤维细胞的增殖速度快于正常大鼠 ,α1(Ⅰ )前胶原的mRNA表达及蛋白表达也高于正常大鼠。结论 糖尿病心肌成纤维细胞在糖尿病心肌病变中增殖加速、胶原合成增加 。
Purpose. To study the effects of myocardial fibroblasts on fibrosis in diabetic cardiomyopathy. Methods. Ten streptozotocin induced diabetic rats were raised for 12 weeks. Then 2 × 107 myocardial fibroblasts were taken out for primary culture. We counted the numbers of cultured cells to observe their growth. RT-PCR and western blot analyses were used to show mRNA and protein expression of pro α1(I) collagen. Results. Proliferation rate of myocardial fibroblasts in diabetes rats was higher than that of myocardial fibroblasts in normal rats. So was mRNA and protein expression of pro α1(I) collagen. Conclusions. Myocardial fibroblasts in diabetic rats accelerate the proliferation of themselves and the synthesis of collagen in diabetic cardiomyopathy. Myocardial fibroblasts take great part in fibrosis in diabetic cardiomyopathy.
