摘要
目的 观察酪氨酸蛋白激酶抑制剂金雀异黄素(genistein)对 GRC- 1细胞的体外生物学行为的影响 ,探讨蛋白酪氨酸激酶 (PTK)抑制剂应用于肿瘤治疗的可能 .方法 将细胞用金雀异黄素处理后 ,分别用 MTT法、激光共聚焦显微镜、电子显微镜观察金雀异黄素对肿瘤细胞的体外生物学行为的影响 .结果 金雀异黄素可显著抑制 GRC- 1细胞的增殖 ,使肿瘤细胞增殖停滞在 G1期和 G2期 ,并明显改变细胞的外在形态和细胞内波形蛋白的排列方式 .结论 酪氨酸蛋白激酶抑制剂 genisein可抑制肾肿瘤细胞的增殖 (P<0 .0 1) ,并改变其细胞骨架的排布方式 .酪氨酸激酶抑制剂有望开发成为有效的抗恶性肾肿瘤的药物 .
AIM To investigate the effect of genistein on the in vitro biological behavior of kidney carcinoma cell line GRC 1 and explore the possibility of developing protein tyrosine kinase inhibitor as a kind of anti cancer drug. METHODSAfter exposing the kidney carcinoma GRC 1 cells to 20 μmol·L -1 and 40 μmol·L -1 genistein, MTT method, electron microscope and laser confocal microscopy are employed to observe the effect of genistein on the in vitro biological behavior of GRC 1 cells. RESULTS Genistein may drastically inhibit the proliferation of GRC 1 cells and stagnate this process in the G1 and G2 phases and also obviously change the outline form and the arrangement of cytoplasmic vimentin. CONCLUSION Genistein can inhibit the proliferation of renal cell carcinoma cells and affect the arrangement pattern of cell skeleton. The protein tyrosine kinase inhibitor may be developed as a new kind of effective kidney neoplasm inhibitor .
