摘要
五种兽用氟喹诺酮药物在体外对猪链球菌、大肠杆菌均具有不同程度的抗菌后效应 (PAE) ,最长者可达3 2 3h ,且PAE受抗菌药物的种类及其浓度、细菌种类及接种量、药物与细菌的接触时间的影响。研究通过在猪的皮下埋植组织笼建立组织笼感染模型 ,成功的应用于沙拉沙星对猪链球菌、猪大肠杆菌的体内PAE研究。结果表明 ,3种浓度的沙拉沙星在体内对 2种细菌均获得了较体外长得多的PAE(P <0 0 1) ,其中对猪链球菌最长的体内PAE值达 3 84h ,对猪大肠杆菌最长的体内PAE值达 2 87h。
Postantibiotic effects(PAEs),the persistent suppression of bacterial growth following a brief exposure to an antibiotic,of five fluoroquinolones for veterinary use against two pathogens of pig in vitro were described.At the same time,several factors influencing the duration of PAE,such as the type of antimicrobial agent and organism,inoculum,concentration of antimicrobial,duration of exposure to antimicrobial were investigated.The PAEs of five agents against two bacteria were induced following exposure to the concentration equal to 2,5,8 times the MICs of these agents in Mueller-Hinton broth at 37℃ for 0.5,1.0,2.0 h,Bacterial regrowths were determined by measuring the viable counts after antibiotic elimination by a 10 -2 dilution procedure.All of the agents exhibited a certain degree of PAE and the longest was demonstrated 3.23 h for difloxacin against E.coli. At the same time,the infected tissue cage model in pigs with normal host defence mechanism had been developed successfully using subcutaneously implanting two plastic tissue cages in each animal.This model was used to determine PAEs of sarafloxacin against S.suis and E.coli in vivo after exposure to the drug for 1 h.The results showed that PAE's duration of sarafloxacin in vivo were longer significantly than that in vitro(P <0.05).The PAEs of sarafloxacin prolonged 3.84 h against S.suis and 2.87 h against E.coli in vivo .
