摘要
用电击法取 5龄第 4天的家蚕肠液 ,加入家蚕核多角体病毒 (BombyxmoriNucleopolyhedrosisVirus)和宿主域扩大的苜蓿尺蠖核多角体病毒 (HybridAutographacalifornicaNucleopolyhedrosisVirus)的多角体或游离病毒粒子 ,分别作用 70min和 4 0min后 ,感染家蚕细胞Bm 5和秋粘虫细胞Sf 2 1,发现病毒粒子已经被家蚕肠液灭活。进一步用BmNPV多角体喂饲 2龄蚕后 ,收集 2 4h内的蚕粪 ,用培养基浸泡后 ,抽提液中亦不含有具感染性的病毒粒子。上述结果说明家蚕肠液对昆虫杆状病毒的病毒粒子具有很好的灭活作用。因而 ,在家蚕核多角体病毒病预防上 ,提高蚕的健康水平、注意起蚕处理和饲养密度、及早发现和隔离病蚕是有效的防治措施。
Polyhedron derived virus(PDV) and budded virus (BV) from Bombyx mori Nucleopolyhedrosis virus (BmNPV) or host rang expanded Hybrid Autographa californica Nucleopolyhedrosis virus (HyNPV) were treated respectively with intesinal digestive juice collected by electric stimulus method of silkworm at fourth day in fifth instar.After infecting Bm 5 and Sf 21 cell line for 70 and 40 minutes separately,it was found that both PDV and BV were inactivated.Feces from 2nd instar larvae which had been fed on BmNPV polyhedra for 24 hours were extracted with TC 100 basal medium.The noninfection of insect cells with the extract revealed as well that virons contained in polyhedra were inactivated by the digestive system.The results suggest that the intestinal digestive juice had a good inactivation to baculoviruses.So,in the prevention of silkworm Nucleopholyhedrosis,these are efficient measures to improve the health level of the silkworm larvae,pay attention to the treatment after ecdysis and rearing density,find and isolate the sick silkworm in early stage.
