摘要
目的 探讨+ GZ作用后,大鼠心肌组织内内皮素-1(ET-1)和内皮型一氧化氮合酶(eNOS)m RNA 表达变化。 方法 24只雄性Wistar 大鼠随机分成对照组、+ GZ重复暴露后30 m in、6 h 和24 h四组,每组6只。对照组大鼠G 值为+ 1 GZ, 实验组大鼠在动物离心机上经历了3次+ 10 GZ/3 m in(两次中间间隔30 m in)作用。分别于暴露后30 m in、6 h 和24 h 处死大鼠取心,提取总RNA,用半定量反转录聚合酶链反应(RT-PCR)检测方法检测大鼠心肌组织ET-1和eNOSm RNA 表达水平。 结果 + GZ暴露后30 m in 和6 h 大鼠心肌组织ET-1 m RNA 明显升高,eNOSm RNA 明显降低,但24 h 时均恢复正常。 结论 + GZ暴露可使大鼠心肌组织内ET-1和eNOSm RNA 表达发生改变,其在心肌损伤中可能起一定的作用。
To explore the changes of mRNA expression of endothelin 1 (ET 1) and endothelial nitric oxide synthase (eNOS) in the heart of rat exposured to repeated +G Z. Methods Twenty four concious Wistar rats served as subjects. They were randomly divided into 4 groups. Using an animal centrifuge, control rats ( n =6) were exposed to +1 G Z and experimental rats ( n =18) were exposed to +10 G Z three times, each for 3 min with 30 min interval in between. The hearts were taken 30 min, 6h and 24 h after the last centrifuge run and total RNA was isolated by single step acid guanidinium thiocyanate phenol chloroform extraction method. The mRNA expression levels of ET 1 and eNOS in rat heart were measured by semi quantitative reverse transcription polymerase chain reaction (RT PCR). Results The ET 1 mRNA expression levels in rat hearts taken 30 min and 6 h after repeated +G Z exposures were significantly higher than those of control group, the eNOS mRNA levels were significantly lower, but the differences between the levels of control group and those of experimental rat heart taken 24 h after +G Z exposure were non significant. Conclusion It is suggested that ET 1 and eNOS mRNA expression in rat heart can be altered by repeated +G Z exposures and the altered expression of ET 1 and eNOS may play a role in the pathologic course of heart damage induced by+G Z exposures, but the changes were reversiable.
