新城疫抗体血凝抑制试验比对结果分析 被引量:1

摘要 为掌握某兽医实验室新城疫抗体血凝抑制(HI)试验能力,用新城疫标准阳性血清和阴性血清制备52份盲样,由6名检测技术员同时进行HI抗体检测。将检测结果与参考值进行比较,计算阳性、阴性一致性以及符合率,并用Win Episcope 2.0软件进行Kappa一致性检验,计算二者差值并进行统计描述。结果表明,312个检测值阳性率为13.5%,低于参考值15.4%的阳性率(p<0.01);二者阳性结果的一致性为68.8%(33/48),阴性结果一致性为96.6%(255/264),符合率为92.3%(288/312),Kappa值为0.69(95%置信区间0.58~0.80);检测值与参考值差值在2个滴度范围内浮动(95%置信区间-2.2~1.8),完全符合的占70.5%(220/312)。由此可知,该兽医实验室新城疫抗体HI试验定性结果与参考值一致性强度为高度,但在定量结果方面仍有进一步提升的空间。 A comparison of HI method for detection of Newcastle Disease(ND)antibody was conducted to estimate the test capability of HI methodat a veterinary laboratory.The standard NDV antibody positive serum which was diluted into different titers,together with the standard NDV-antibodynegativeones were prepared totally 52 for the 6 technicians who did the HI test at the same time.The HI titers of the 52 samples were observed by thestaffs and were compared with the reference values.The positive and negative coincidence,the coincidence rate were calculated separately and Kappatestwas conducted by using Win Episcope 2.0 software. Meanwhile,difference between the tested and the reference values was described by statisticsmethod.The results showed that positive rate was 13.5%,which showed a statistic difference with the reference 15.4%(p约0.01).The positive consistencyrate was 68.8%(33∕84),the negative consistency rate was 96.6%(255∕264)and the coincidence rate was 92.3%(288/312).The Kappa value was 0.69 (95% CI= 0.58-0.80).The difference between the observed and the reference values was existed,and the different values were at the range of -2.2 to1.8(95% CI),in which the rate of fully agreement of these two values was 70.5%(220∕312).The results indicated that,for the qualitative test of NDVHI antibody there was a high agreement between the observed results and the reference values at this veterinary laboratory,but the quantitative testshowd be further improved.
