蒲公英炮制前后总黄酮含量差异研究 被引量:9

摘要 目的采用紫外分光光度计法测定蒲公英炮制前后总黄酮的含量的变化,为临床上采用蒲公英及蒲公英炭治疗胃炎、胃溃疡的效果提供科学依据。方法以乙醇浓度,超声时间,超声温度,超声功率为影响因素,设计正交试验L9(34)对蒲公英及蒲公英炭中的黄酮进行提取;用紫外分光光度计测定总黄酮含量并确定最优超声方法;并比较蒲公英及蒲公英炭的总黄酮含量。结果在提取溶剂为65%乙醇,超声功率为462 W,超声时间为35 min,超声温度为60Ⅰ的条件下,蒲公英与蒲公英炭提取率最高,分别为3.9%和2.9%;蒲公英及蒲公英炭中芦丁在0.017 2~0.062 7 mg/mL具有良好的线性关系,回归方程为C=0.091A-0.001,r=0.997 5;蒲公英供试品溶液与蒲公英炭供试品溶液的稳定性RSD分别为0.42%和0.60%;重复性RSD分别为0.85%和0.70%,精密度为1.04%,表明该工艺稳定可行,准确性高。结论蒲公英炮制后其总黄酮的含量明显小于蒲公英原药材,就总黄酮含量这一指标来说,临床上治疗胃炎、胃溃疡应优先考虑选用蒲公英原药材。 Objective This experiment by ultraviolet spectrophotometer method, the change of the content of total flavonoids determination of dandelion before and after processing, for clinical using dandelion raw medicinal herbs to treat gastritis, gastric ulcer or dandelion charcoal will provide a scientific basis for treatment of gastritis, gastric ulcer. Methods Ethanol concentration, ultrasonic time, ultrasonic temperature, ultrasonic power for influence factors, the design of orthogonal test L9(34) of dandelion extract and flavonoids of dandelion carbon; Flavonoids content calculated to determine the optimal ultrasonic method; And the optimal method of ultrasonic extraction of dandelion and a comparative study on flavonoids content of dandelion carbon. Results The extraction solvent was 65% ethanol, ultrasonic power is 462 W, ultrasonic time is 35 min, ultrasonic temperature is 60℃ under the condition of ultrasonic, dandelion and dandelion charcoal withdrawal rate is highest, at 3.9% and 2.9% respectively; Dandelion and rutin in dandelion carbon at 0.017 2 - 0.062 7 mg/mL has A good linear relationship, the regression equation for C = 0.091 A-0.001, r = 0.997 5; Dandelion and dandelion carbon of test sample solution stability of test sample solution RSD were 0.42% and 0.60% respectively; Repeatability RSD were 0.85% and 0.70% respectively, the precision is 1.04%, show that the process stability is feasible and high accuracy. Conclusion Dandelion processing after the flavonoids content is much smaller than the dandelion, total content of flavonoids as the index for the clinical treatment of gastritis, gastric ulcer priority should be given to choose the dandelion raw medicinal materials.
