摘要
目的:研究中国南方汉族人群中微小RNA-146a(miR-146a)rs2910164多态性与急性冠脉综合征(acute coronary syndrome,ACS)的关系。方法:选择急性ACS患者100例,以冠状动脉正常的患者作为对照(n=100),常规检测血压、体重指数、空腹血糖、血脂、肝肾功能等,PCR检测miR-146a rs2910164基因的多态性,观察CC、GG和CG不同基因型以及该位点C、G不同等位基因频率在ACS患者和正常冠状动脉人群中的分布情况。结果:在ACS患者中,miRNA-146a rs2910164位点多态性的次要C等位基因频率显着高于对照组(OR=0.599,95%CI=0.398~0.900,P=0.013)。ACS患者的GC基因型(OR=0.480,95%CI=0.260~0.887,P=0.018)和CC基因型(OR=0.399,95%CI=0.164~0.971,P=0.040)的频率显著高于GG基因型频率(χ^(2)=0.07,P=0.791),两组之间的差异具有统计学意义(P<0.05)。与GG基因型比较,杂合子GC基因型在BMI(P=0.017)、FBG(P=0.022)、TG(P=0.009)、TC(P=0.001)、LDL(P=0.001)、收缩压舒张血压(P=0.001)均显著升高。CC基因型也表现出更高的BMI(P=0.035)、FBG(P=0.018)、TG(P=0.013)、TC(P=0.001)、LDL(P=0.001)、收缩压和舒张压(P=0.001),差异有统计学意义(P<0.05)。结论:miRNA-146a rs2910164的C等位基因与中国南方汉族人群ACS及其表型的易感性增加呈正相关。
Objective To investigate the association between the polymorphism of microRNA-146a rs2910164 gene and acute coronary syndrome(ACS)in a southern Chinese Han population.Method 100 patients with ACS and 100 patients with normal coronary arteries were enrolled,whose blood pressure,body mass index(BMI),blood fat,fasting plasma glucose(FBG),serum creatinine were routinely measured.Performed by polymerase chain reaction(PCR),the genoUpes of miR-146a rs2910164 were measured and the frequency of C,G allele were investigated.Results The C allele had a significantly higher fre-quency in ACS patients(OR=0.599,95%CI=0.398-0.900,P=0.013).The frequencies of GC(OR=0.480,95%CI=0.260-0.887,P=0.018)and CC genoUpes(OR=0.399,95%CI=0.164-0.971,P=0.040)were significantly higher than the frequency of the GG genoUpe in ACS patients(χ^(2)=0.07,P=0.791).The heterozygote GC and the rare CC genoUpes showed a significant increase in body mass index(P=0.071,P=0.035),fasting blood glucose(P=0.022,P=0.018),tri-glycerides(P=0.009,P=0.013),total cholesterol(P=0.001,P=0.001),low-density lipoprotein(P=0.001,P=0.001),systolic and diastolic blood pressure(P=0.001,P=0.001),the difference was statistically significant(P〈0.05).Conclusion The C allele of miRNA-146a rs2910164 showed positive association with increased susceptibility to ACS and its phenotypes in the study population.
