摘要
目的分别观察硫氢化钠(Na HS)对低糖和高糖环境下的人肾小管上皮细胞株HK-2氧化应激的影响。方法传代培养HK-2用无血清培养基同步化24 h后,将各组细胞分为8组:低糖对照组(5. 5 mmol/L D-葡萄糖)、低糖+不同剂量Na HS组(50、100、200μmol/L)、高糖组(40 mmol/L D-葡萄糖)、高糖+不同剂量Na HS组(50、100、200μmol/L)。将细胞置于37℃恒温培养箱中刺激培养24、48 h,收集细胞和培养液。比色法测定培养液中SOD活性、MDA含量、GSH-px活力和H2O2水平,激光共聚焦观察ROS水平。结果与低糖对照组比较,高糖可明显降低肾小管上皮细胞培养液中SOD活性和GSH-px活力(P <0. 01),增加MDA含量(P <0. 05)、H2O2水平(P <0. 05)以及肾小管上皮细胞内ROS的产生。不同浓度Na HS可提高低糖和高糖肾小管上皮细胞培养液中SOD活性和GSH-px活力(P <0. 05),降低MDA含量(P <0. 05)、H2O2水平(P <0. 01)以及肾小管上皮细胞内ROS的产生。其中,均以Na HS中剂量100μmol/L变化明显。结论Na HS可能通过降低肾小管上皮细胞内氧化应激而对糖尿病肾病起保护作用。
Objective To investigate the effects of Sodium hydrogen sulfide (NaHS,a donor of H 2S) on oxidative stress by high glucose in human renal tubular epithelial cells. Methods Human renal tubular epithelial cells were cultured and divided into eight groups:low glucose as control group (5.5 mmol/l D-Glucose),low glucose+NaHS pretreatment group with concentration of 50,100 and 200 μmol/l,high glucose group (40 mmol/l D-Glucose),high glucose+NaHS pretreatment group with concentration of 50,100 and 200 μmol/l.The SOD activity,MAD conent,GSH-px activity and H 2O 2 level were determined by colorimetry,and the ROS level was observed by laser. Results Compared with the low sugar control group,high sugar reduced SOD activity and GSH-px activity,increased MAD conent and H 2O 2 level in the culture medium,increased ROS production in renal tubular epithelial cells.NaHS increased SOD activity and GSH-px activity,reduced MAD conent and H 2O 2 level in the culture of low and high sugar renal tubules epithelial cells,decreased ROS level in renal tubular epithelial cells.Among them,the dose 100 μmol/l of NaHS had the best effects. Conclusion NaHS may protect diabetic nephropathy by reducing oxidative stress in renal tubular epithelial cells.
