鼠疫耶尔森菌耐消毒剂基因检测方法的建立与应用 被引量:1

摘要 目的利用聚合酶链式反应(PCR)技术建立鼠疫耶尔森菌(以下简称鼠疫菌)耐消毒剂基因的检测方法,为控制鼠疫感染提供指导性建议。方法根据Gen Bank公布的耐消毒剂磺胺复合物qac EΔl-sull基因序列,设计一对引物,对分离自青海省境内鼠疫自然疫源地的188株野生鼠疫菌的DNA进行PCR扩增,扩增产物进行凝胶电泳,用凝胶成像系统拍照后读取结果。结果阴性对照和阳性对照均成立,所测样本的PCR扩增结果均为阴性,尚未发现具有抗消毒剂基因的菌株。结论利用PCR技术对鼠疫菌进行耐消毒剂基因的检测方法是可行的,该方法能为大批量鼠疫菌的耐消毒剂基因的筛选提供高效、经济且准确率高的实验手段。 Objective To establish the detection method of the disinfectant-resistant gene in Yersinia pestis based on polymerase chain reaction( PCR) technology,so as to provide guidance for the control of plague infection. Methods One couple of primer was designed according to the sequence of the qac EΔl-sul1 gene of disinfectant sulfonamide complex published in Genbank. PCR was performed on 188 wild Yersinia pestis isolates isolated from the natural epidemic of plague in Qinghai Province. The amplified products were subjected to gel electrophoresis. The results were read using a gel imaging system. Results Both the negative control and the positive control were established. The PCR amplification results of the tested samples were all negative. No strains was found with disinfectant-resistant gene. Conclusion It is feasible to use PCR technology to detect disinfectant-resistant gene in Yersinia pestis with efficient,economical and accurate results for screening plenty of strains.
