摘要
目的建立高效液相色谱-串联质谱法检测猪肉及牛奶中15种β-受体激动剂的方法。方法样品经β-葡萄糖醛苷酶酶解,依次用乙酸乙酯及叔丁基甲醚提取,MCX固相萃取柱净化,以0. 1%甲酸水溶液-0. 1%甲酸乙腈溶液作为流动相进行洗脱,用XTerra C18(2. 1 mm×150 mm,3. 5μm)色谱柱分离,采用多反应监测(MRM)模式进行定性和定量分析。结果 15种β-受体激动剂线性相关系数均≥0. 992,平均回收率为76. 5%~119. 6%,相对标准偏差为1. 0%~12. 8%(n=6)。结论该方法简单、快速、灵敏,可用于猪肉及牛奶中15种β-受体激动剂的检测。
Objective To establish a method for the determination of 15 β-agonists residues in pork and milk by high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS). Methods The samples were enzyme hydrolyzed with β-glucuronidase sulfatase,and then the analytes were extracted with ethyl acetate and methyl tert-butyl ether,and cleaned up on a MCX cartridge,and finally separated on a C18 column(150 mm × 2. 1 mm,3. 5 μm) by using acetonitrile(containing 0. 1% formic acid) and water(containing 0. 1% formic acid) as mobile phase. The Qualitative analysis and quantitative analysis were carried out in multiple reaction monitoring(MRM) model. Results The correlative coefficient of 15 β-agonists were ≥0. 992. The average recoveries were within 76. 5%-119. 6%,and the relative standard deviations ranged from1. 0% to 12. 8%(n = 6). Conclusion The method is simple,rapid and sensitive. It is suitable for the simultaneous determination of 15 β-agonists in pork and milk.
