摘要
目的探讨白及多糖对PM2.5致急性肺损伤的干预作用。方法水提醇沉法制备白及多糖,无机法提取PM2.5并制成悬液。ICR小鼠以不同浓度的白及多糖(5 mg/ml、20 mg/ml、40 mg/ml)预保护给药1周后,连续气管滴注PM2.5悬液(0. 4 mg/40μl/只)急性染毒3 d,建立预保护模型组;连续气管滴注PM2.5悬液(0. 4 mg/40μl/只)急性染毒3 d,造模期间持续给药不同浓度的白及多糖(5 mg/ml、20 mg/ml、40 mg/ml),建立治疗模型组。观察肺组织形态学变化,采用流式液相多重蛋白定量技术检测肺泡灌洗液炎症因子蛋白含量。结果白及多糖可以有效改善PM2.5致小鼠肺部炎性细胞浸润及呼吸道的黏膜损伤和支气管周围组织的炎性浸润,有效抑制IL-6、IFN-γ、TNF-α、MCP-1炎症因子表达。结论白及多糖通过预防、治疗缓解机体炎症损伤,抑制PM2.5致急性肺损伤作用。
Objective To investigate the intervention effects of Bletilla striata polysaccharide on PM2.5-induced acute lung injury. Methods We prepared the Bletilla striata polysaccharide by water extraction and alcohol precipitation method. PM2.5 was extracted by inorganic method and made into suspension. After one week' s feeding of different concentrations of Bletilla striata polysaccharide(5 mg/ml,20 mg/ml,40 mg/ml) with pre-protection,mice underwent tracheal instillation of PM2.5 suspension(0. 4 mg or 40 μl for 1 mouse) for three days to establish a pre-protection model group. Continuous tracheal instillation of PM2.5 suspension(0. 4 mg or 40 μl for 1 mouse) was performed for three days,during which different concentrations of Bletilla striata polysaccharides(5 mg/ml,20 mg/ml,40 mg/ml) were continuously administered to establish a treatment model group. Morphological changes of lung tissue were observed. Cytokine expression in BALF was detected by CBA method.Results Bletilla striata polysaccharide could effectively improve the lung inflammatory cell infiltration and respiratory tract mucosal injury and inflammatory infiltration of bronchial tissue in PM2.5-induced model mice. Bletilla striata polysaccharide can effectively inhibit the expression of inflammatory cytokines of IL-6,IFN-γ,TNF-α,MCP-1. Conclusion Bletilla striata polysaccharide can relieve the inflammation of the body and inhibit the toxicity of PM2.5 to acute lung injury by preventing and treating the inflammatory response of the body.
