摘要
建立了高效液相色谱紫外检测器测定浓缩磷脂软胶囊中磷脂酰胆碱的测定方法。采用Sino Chrom柱(I.D.4.6 mm×250 mm, 5μm),在柱温为45℃,流动相为异丙醇-正己烷-水(70︰16︰14, V/V),流速为1.2 mL/min,检测波长为205 nm的条件下进行测定。结果显示,在0.2~2.0 mg·mL^(-1)浓度范围内线性关系良好,相关系数大于0.999 6,添加水平的回收率平均值为98%, RSD为2.41%。该方法在对磷脂酰胆碱的测定上具有操作简便、专属性强、准确度高、成本低的特点,适用于浓缩磷脂软胶囊的质量控制。
It was to establish an effective method to determine phosphatidylcholine in concentrated phospholipid soft capsule by high performance liquid chromatography UV detector. The Sino Chrom column （I.D. 4.6 mm×250mm, 5 μm） was adopted in 45 ℃ with the flow phase of Visopropanol： Vhexane： Vwater=70 ： 16 ： 14, 1.2 mL/min velocity, and 205 nm detection wave length.The results showed that it was well correlated in linear range of 0.2-2.0 mg.mL^-1, correlation coefficient was above 0.999 6, average recovery rate was 98%, and the relative standard deviation was 2.41%. The method was suitable for the determination of phosphatidylcholine with simple operation, high specificity, high accuracy and low cost, and it could be applied to quality control of concentrated phospholipid soft capsules.
