摘要
目的:探讨辛温通阳中药对急性心肌缺血大鼠模型的保护作用及机制。方法:70只雄性SD大鼠随机分为假手术组、模型组、薤白组、附子组、干姜组、桂枝组、硝酸甘油组,每组10只。采用结扎左冠状动脉制备心肌缺血大鼠模型。造模完成后给药取材,观察各组大鼠心电图改变、心肌病理形态及氧化应激、炎症因子等指标。结果:与假手术组比较,模型组ST段明显抬高;与模型组比较,治疗组大鼠ST段降低;HE染色显示,假手术组心肌组织结构完整,模型组心肌纤维排列紊乱,心肌纤维断裂、肿胀,治疗各组心肌纤维排列相对整齐,部分心肌纤维断裂;与假手术组比较,模型组总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px)活性及白介素-10 (IL-10)水平明显降低,丙二醛(MDA)、白介素6(IL-6)、肿瘤坏死因子(TNF-α)水平及髓过氧化物酶(MPO)活性显著升高,与模型组比较,药物组T-AOC、GSH-Px活性及IL-10水平升高,MDA、IL-6、TNF-α水平及MPO活性降低(P<0.05,P<0.01)。结论:辛温通阳中药能够改善急性心肌缺血模型大鼠的缺血状态,对心肌缺血具有保护作用。
Objective ： To explore the protective effect and mechanism of activating yang with herbs acrid in fla- vor and warm in property on acute myocardial ischemia （AMI） in rats. Methods： 70 male SD rats were ran- domly divided into the sham operation group, the model group, Xiebai （Bulbus Allii Macrostemonis ） group, Fuzi （Radix Aconiti Lateralis Praeparata ） group, Ganjiang （ Rhizoma Zingiberis ） group, Guizhi （ Ramulus Cin- namomi）group, and Nitroglycerin group, with 10 rats in each group. The model was made by ligating left coronal7- artery. The changes of ECG, pathological morphology, oxidative stress, and inflammatol7- factors were observed after intervention. Results： The ST segment was significantly elevated in the model group com- pared to that in the sham operation group; it was lowered in all the medication groups compared to that in the model group. HE staining showed that the myocardium tissue of the sham operation group was intact; however the arrangement of the myocardial fibers in the model group was disordered, and the myocardial fibers were broken and swollen; in all medication groups, the arrangement of the myocardial fibers was relatively neat, and just some of the myocardial fibers were broken. The activities of T - AOC, GSH - Px, and IL - 10 were significantly decreased and the levels of MDA, IL - 6, TNF - or, and MPO were significantly increased in the model group compared to those in the sham operation group ; the activities of T - AOC, GSH - Px, and IL - 10 were increased and the levels of MDA, IL -6, TNF -or, and MPO were decreased in all the medication groups compared to those in the model group （P 〈 0.05, P 〈 0.01 ）. Conclusion ： Activating yang with herbs acrid in flavor and warm in property can improve the acute myocardial ischemia in rats with its protective effect.
