党参多糖的提取及体内外免疫作用的研究 被引量:26

摘要 目的旨在分离、纯化党参多糖并对其在体内、外的免疫调节活性进行研究。方法采用传统的水提(加热回流)醇沉(梯度乙醇沉淀)法制备党参粗多糖(Codonopsis pilosula Polysaccharide,CPP),经活性炭吸附脱色、Sevage法除蛋白得党参多糖,通过体外淋巴细胞增殖实验检测CPP的免疫调节作用,通过测定大鼠免疫脏器指数、血清中补体C3、补体C4、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、免疫球蛋白A(IgA)、生长激素(GH)、胰岛素样生长因子1(IGF-1)、三碘甲状腺原氨酸(T3)、甲状腺素(T4)、胰岛素(INS)生成水平评价其免疫调节作用。结果 CPP能够促进淋巴细胞体外增殖(P<0.05);党参多糖各剂量组均能提高胸腺指数和脾脏指数,与模型组相比,党参多糖高、中、低剂量组的血清中C3、C4、IgG、IgM、IgA、GH、IGF-1、T3、T4、INS生成水平均有不同程度的增高(P<0.05)。结论 CPP对环磷酰胺所致免疫功能低下大鼠具有明显的增强作用,本研究为CPP的临床应用和功能性食品的开发提供了理论依据。 Objective The purpose is to isolate and purify Codonopsis pilosula Polysaccharide and to study its immunomodulatory activity in vitro and in vivo.Methods Codonopsis pilosula Polysaccharide(CPP)was prepared by traditional water-extraction(heated reflux)alcohol precipitation(gradient ethanol precipitation)method.The activated carbon was decolorized by adsorption and the polysaccharide was removed by Sevage method.Result The proliferation of Codonopsis pilosula Polysaccharide CPP immunosuppressive effect by measuring the index of rat immune organ,serum complement C3,complement C4,immunoglobulin G(IgG),immunoglobulin M(IgM),immunoglobulin A(IgA),growth hormone GH,IGF-1,T3,T4,INS were measured to evaluate the immunoregulatory effects.Results CPP could promote the proliferation of lymphocytes in vitro(P〈0.05).Compared with the model group,the levels of C3,C4,The levels of IgG,IgM,IgA,GH,IGF-1,T3,T4 and INS were all increased to a certain extent(P〈0.05).Conclusion CPP can significantly enhance the immunosuppressive mice induced by cyclophosphamide This study provides a theoretical basis for the clinical application of CPP and the development of functional foods.
