肌醇多磷酸5磷酸酶PIPP在小鼠受精卵内对细胞周期蛋白Cdc2的影响 被引量:2

摘要 目的研究脯氨酸丰富的肌醇多磷酸5磷酸酶(PIPP)在小鼠1-细胞期胚胎中的定位以及它对Cdc2在Tyr15位点磷酸化的调节作用。方法提取质粒、酶切鉴定并转染PEFBOS PIPP质粒以及空载体PEFBOS vector到小鼠受精卵,用免疫荧光染色检测PIPP的定位,Western blot检测PIPP对Cdc2在Tyr15位点磷酸化的影响。结果PIPP的定位在小鼠受精卵有丝分裂过程中不断变化,并且PIPP在小鼠受精卵中的过度表达增加了Cdc2在Tyr15位点的磷酸化。结论 PIPP存在于小鼠受精卵中并通过调节Cdc2的磷酸化参与小鼠受精卵的有丝分裂过程。 Objective To study the effect of proline-rich inositol polyphosphate 5-phosphatase(PIPP) in mouse fertilized eggs on cell cycle protein Cdc2.Methods PIPP was observed in mouse fertilized eggs by indirect immunofluorescent staining.The phosphorylation of Cdc2 Tyr15 was detected by Western blot,and transfecting to mouse fertilized eggs of PEFBOS PIPP and PEFBOS vector plasmids.Results The distributions of PIPP were observed in cell membrane of G1 phase and whole cell of M phase in mouse fertilized eggs.And the overexpression of PIPP increased Cdc2 phosphorylation at Tyr15.Conclusion PIPP exists in mouse fertilized eggs and participates in mitotic regulation in mouse fertilized eggs via Cdc2.
