摘要
目的观察医用臭氧水对兔膝关节骨性关节炎关节软骨基质金属蛋白酶1/3/13(MMP-1/3/13)表达的影响。方法成年新西兰大白兔36只,随机分为对照组12只、骨性关节炎组12只与医用臭氧水治疗组12只。采用Hulth法将骨性关节炎组与医用臭氧水治疗组实验动物制成膝关节骨性关节炎模型。待造模成功后,医用臭氧水治疗组向兔膝关节腔内注射医用臭氧水2ml,注射浓度为20μg/ml,一周一次,共治疗3次,骨性关节炎组兔关节腔内注射空气;对照组实验动物不作处理。在造模成功时和处死前测定兔膝关节活动度,末次干预1周后,用空气栓塞法处死实验兔,取下实验兔右膝关节软骨进行组织匀浆,采用Western blot与real time PCR方法测定关节软骨中MMP-1、MMP-3与MMP-13的含量变化。结果与对照组相比,造模成功后的骨性关节炎组和医用臭氧水治疗组兔膝关节活动度显著降低,P<0.01;与骨性关节炎组相比,采用医用臭氧水治疗后,医用臭氧水治疗组兔膝关节活动度显著升高,P<0.01。骨性关节炎组大鼠膝关节软骨组织中MMP-1、MMP-3与MMP-13的蛋白与m RNA表达水平比对照组明显升高,而医用臭氧水治疗组比骨性关节炎组显著降低,P<0.01。结论医用臭氧水对骨性关节炎的治疗作用可能与降低早期骨性关节炎关节软骨中MMP-1、MMP-3与MMP-13的表达水平相关。
Objective To observe the effect of medical ozone water on the expressions of matrix metalloproteinase-1/3/13(MMP-1/3/13) in articular cartilage of knee osteoarthritis rabbits.Methods A total of 36 New Zealand white rabbits were randomly divided into control group,osteoarthritis group(established by Hulth method)and medical ozone water treatment group(20 g/2 ml medical ozone water,injected into knee joint cavity,once a week,for 3 times in total).All the rabbits were killed with air embolism 1 week after the last intervention,and then the cartilage of the right knee was removed,tissue homogenates were taken.The expressions of MMP-1/3/13 proteins and m RNA were detected by Western blot and real time PCR methods respectively.Results The activity degree of knee joint was significantly decreased in osteoarthritis group than in the control group,but increased in medical ozone water treatment group than in osteoarthritis group.The expression levels of MMP-1/3/13 proteins and m RNAs were significantly increased in osteoarthritis group than in the control group,but decreased in medical ozone water treatment group than in osteoarthritis group.Conclusion The therapeutic effect of medical ozone water on knee osteoarthritis might be related to the downregulation of MMP-1/3/13 expressions in articular cartilage of osteoarthritis.
