摘要
目的探讨HIF-1α的表达程度对人骨肉瘤细胞自噬的影响。方法传代培养人骨肉瘤细胞株(MG-63),施加不同条件诱导各组细胞中HIF-1α不同程度表达,MTT法检测细胞增殖抑制率变化,Western blot检测HIF-1α、Beclin-1、LC3Ⅱ/LC3Ⅰ表达。结果低氧条件下HIF-1α表达增加,细胞增殖受到抑制,同时LC3Ⅱ/LC3Ⅰ,Beclin-1表达水平增加;当诱导HIF-1α过度表达时,细胞增殖受抑制更明显,同时LC3Ⅱ/LC3Ⅰ,Beclin-1表达水平增加亦更明显;当HIF-1α表达受到抑制时,细胞增殖受抑制减弱,同时LC3Ⅱ/LC3Ⅰ,Beclin-1表达水平亦较少。结论 HIF-1α可能参与人骨肉瘤细胞的发病机制。
Objective To investigate the effect of HIF-expression on autophagy of human osteosarcoma cells.Methods Human osteosarcoma cells(MG-63) were cultured in different conditions,and the degree of autophagy was detected by MTT and the expression of autophagy related proteins were detected by Western blot.Results Under hypoxic conditions,HIF-1α expression level was increased,cell proliferation was inhibited,while LC3 II/LC3 II,Beclin-1 expression levels were increased.While induced by HIF-1α alpha overexpression,cell proliferation was inhibited obviously,but LC3 II/LC3 II,Beclin-1 expression levels were increased more obviously.While HIF-1α expression was inhibited,the proliferation of the cells was inhibited,LC3 II/LC3 II and Beclin-1 expression levels were downregulated.Conclusion HIF-1α might be involved in the pathogenesis of human osteosarcoma cells.
