摘要
目的 :对比观察尼群地平衍生物Ⅵ (NITDⅥ )及尼群地平 (NIT)对蟾蜍离体心脏活动的影响。方法 :取蟾蜍 40只 ,采用离体蛙心灌流实验方法 ,分别观察NITDⅥ及NIT对心肌收缩力和自律性的作用。结果 :NITDⅥ 3× 1 0 - 8mol·L- 1 ~ 3× 1 0 - 5mol·L- 1 可明显抑制蟾蜍离体心脏的收缩力 (P <0 .0 1 ) ,并表现有剂量依赖性 ;NITDⅥ 3× 1 0 - 5mol·L- 1 可同时降低自律性 (P <0 .0 5) ;NIT 3× 1 0 - 9mol·L- 1 ~ 3× 1 0 - 5mol·L- 1 可显著抑制蟾蜍离体心脏的收缩力 ,从 3× 1 0 - 8mol·L- 1 开始明显抑制自律性。结论 :NITDⅥ与NIT对心肌收缩力及自律性均表现有抑制效应 ,但NITDⅥ的抑制作用弱于NIT。
Aim:To observe the effects of nitrendipine derivative Ⅵ(NITD Ⅵ) and nitrendipine (NIT) on isolated heart activity of toads. Methods:Isolated frog heart infusing method was used with 40 toads. Effects of NITD Ⅵ and NIT on cardiac muscles contractive force and automatic rhythmicity were observed.Results:The result showed that NITD Ⅵ obviously inhibited the contractive force of isolated hearts ( P <0.01) at the concentration ranging from 3×10 -8 mol·L -1 to 3×10 -5 mol·L -1 in a dose dependant manner; NITD Ⅵ could decrease automatic rhythmicity at 3×10 -5 mol·L -1 ( P <0.05 );NIT could obviously inhibit the contractive force at the concentration ranging from 3×10 -9 mol·L -1 to 3×10 -5 mol·L -1 , and began to inhibit automatic rhythmicity at 3×10 -8 mol·L -1 . Conclusion: Both NITD Ⅵ and NIT could inhibit cardiac muscles contrative force and automatic rhythmicity, but the effects of NITD Ⅵ are weaker than that of NIT.
